Objective:To investigate the gene expression of11β-HSD1, H6pd and Nr3cl in the skeletal muscle by establishing mouse model of IVF for the exploration of the possible mechanisms of alteration of metabolism in IVF-ET female offspring.Materials and methods:1. Mouse models of IVF and in vivo fertilization were established by using C57BL/6J mice.2. The levels of11β-HSD1, H6pd and Nr3c1mRNA in the skeletal muscle of IVF female mice were detected at3wk,8wk, and1.5yr by RT-PCR.3. The11β-HSD1protein expression in the skeletal muscle of IVF female mice was detected at3wk,8wk, and1.5yr by western blotting. Result (s):1. A total of15IVF and15in vivo mice were delivered.2. Effects of IVF on the expression levels of11β-HSD1H6pd and Nr3cl in the skeletal muscle(1) Expression levels of11β-HSD1and H6pd in the skeletal muscle showed difference between IVF mice and in vivo mice, and there was no difference in gene expression of Nr3c1between the two groups from3wk to old age.(2) At3wk, there was no significant difference in gene H6pd expression of11β-HSD1and between IVF mice and in vivo mice. At8wk, H6pd expression in IVF mice appeared significantly increasing,11β-HSD1expression remains no significant difference. However, when the mice reached to their old age, the significantly higher expression levels in genes of11β-HSD1and H6pd were detected in IVF mice. There was no difference in gene expression of Nr3c1between the two groups from3wk to old age.3. Effects of IVF on the11(3-HSD1protein expression in the skeletal muscleThere was no statistically significant difference in11β-HSD1protein expression between IVF and in vivo groups at other stages.Conclusion (s):1. IVF could affect genes expression of11β-HSD1and H6pd in the skeletal muscle of IVF female mice.2. IVF has no affect on gene expression of Nr3cl in the skeletal muscle of IVF female mice.
