Objective:Recording sciatic nerve discharge when acupuncture in the naive group and SNI model group’s Zusanli(ST36).To compare the influence of acupuncture on sciatic nerve discharges between physiological and pathological states.Methods:Using VonFrey filaments to select the pain thresholds of Sprague Dawley rat.Rat with similar pain thresholds were randomly divided into naive group、naive+acupuncture group、model group.Model group with SNI model caused by the way of Decosterd and Woolf’s were randomly divided into model group and model+acupuncture group.Five days after surgery,recording the sciatic nerve discharge of every group by BL420s.After the electrical signals stability, naive+acupuncture group and model+acupuncture group were given acupuncture for15minutes at ST36with hand-manipulation about1minute and retaining needle about4minutes, repeat three times. Naive group and model just recording the sciatic nerve discharge for15minutes.Then use Clampfit to analyze the frequency and the amplitude’s change rate in each group.Results:1. Five days after surgery,the pain thresholds decreased immediately (p<0.01).2. Recording sciatic nerve discharge by BL420s:(1) Model group:compared with the naive group, frequency is higher than the naive group (p<0.05).(2) The frequency of the naive group increased during acupuncture (p<0.05), the model group’s decreased during acupuncture (p<0.05),the model group’s rate changes of frequency is higher than the naive group (p<0.05).(3) Compared with retaining needle the frequency of the naive+acupuncture group increased during hand-manipulation (p<0.05).Compared with retaining needle the model+acupuncture group’s decreased during hand-manipulation (p<0.05).During hand-manipulation the naive group’s rate changes of frequency is higher than that one when retaining needle (p<0.05).During hand-manipulation the model group’s rate changes of frequency is higher than that one when retaining needle (p<0.05).(4) The changes of amplitude’s change rate in naive group and model group is not significant (p>0.05).(5) The amplitude’s change rate and the amplitude of the naive+acupuncture group increased during acupuncture (p<0.05).The mode+acupuncture group’s amplitude decreased but the amplitude’s change rate during acupuncture increased (p<0.05),the model+acupuncture group’s rate changes of frequency is higher than the naive+acupuncture group (p<0.05).(6) Compared with retaining needle the amplitude of naive+acupuncture group has an increasing tendency during hand-manipulation, rate changes of amplitude is higher than that one when retaining needle (p<0.05). Compared with retaining needle the amplitude of model+acupuncture group has an decreasing tendency during hand-manipulation,but the rate changes of amplitude is not significant (p>0.05).3. Conclusions:1. According to behavioral experiments we conclude that the surgery that made SNI model was successful.2. In different conditions, the sciatic nerve discharge was different during acupuncture.In physiological conditions can induce sciatic nerve discharge,on the opposite side acupuncture inhibit the sciatic nerve discharge in the pathological conditions.and the regulatory capacity in pathological condition was significantly higher than the physiological condition.3. When retaining needle on the acpoint not to manipulate,the nerve discharge was not significant in physiological condition.In pathological condition we can got a better effect to avoid nerve discharge during hand-manipulation.4. According to what we observed may be able to reveal the outer periphery of the mechanism of action of acupuncture.
