Background and Objective:Hypertension which is one of the most important risk factors for cardiovascularsystem diseases such as myocardial infarction, stroke is a global public problem. Thetarget of blood pressure management is no longer confined to the extent ofdecompression. The stationarity and the protection of target organs such asmyocardial cells, blood-vessel, kidney have became an major consideration onscheme selection. Such as JNC8,2014AHA/ACC/CDC and JSH2014, thesehypertension guidelines take the same view without prior consultation: therestatement for an important position of drugs joint application. Sinistral amlodipineand Valsartan were listed and widely used for many years. It’s antihypertensive effecthas been repeatedly clinical validation. However both joint application, especially tothe vascular endothelium protection was seldom reported. In this study, we based on adifferent antihypertensive drug intervention, combined the already known vascularendothelium injury marker and investigated the effect of vascular endotheliumprotection on the combination of sinistral amlodipine and valsartan.Methods:A total of30experimental rats, divided into five groups;24of spontaneouslyhypertensive rats (SHR), sinistral amlodipine were randomly divided into treatmentgroup (group A) and valsartan treatment group (group B), sinistral amlodipine+valsartan treatment group (group C) and blank control group (group D), each group of6;6WKY (SHR control strain) is set to normal control group (group E). Measuringnoninvasive blood pressure and the LVEF%after drug treatment and observation wasgiven8weeks. To dissect myocardial tissue of the executed rats, calculating theHW/BW LVW/BW. The level of eNOS, NADPH, SOD,THF-α,IL-6was detected by Elisa method.Results:1.When group A,B,C compared with group D, NADPH dropped significantly;there isno difference between group A,B and C.2.When group A,B,C compared with group D, SOD increased significantly;whengroup C compared with A,B, SOD increased more.3.When group A,B,C compared with group D, eNOS increased significantly;whengroup C compared with A,B, SOD increased more.4.When group A,B,C compared with group D, IL-6dropped significantly;whengroup C compared with A,B, IL-6dropped more.5.When group A,B,C compared with group D, THF-αdropped significantly;whengroup C compared with A,B, THF-αdropped significantly.6.When group A,B,C compared with group D, HW/BW and LVW/BW droppedsignificantly;when group B,C compared with A,HW/BW and LVW/BW droppedmore.7.On SBP level, group A、B、C dropped in different levels,group D increasedobviously.8.Group D’s LEVF droupped significantly;changes in group A,B,C and E has noclinical significance.Conclusion:1. Sinistral amlodipine and valsartan both have good endothelial protective effect.2. Sinistral amlodipine combines with valsartan is superior to the single drug, whichpossessing synergistic effect on the vascular endothelium protection.
