| Objective:Expression of XIAP, XAF-1and detected by Smac in normal endometrium,atypical hyperplasia endometrium and endometrial carcinoma, in endometrialcarcinoma and explore mechanisms of the three.Methods:2013January to2014December were randomly selected from Jilin University,Department of Obstetrics and gynecology in China Japan Union Hospital, pathologyexamination confirmed the diagnosis of endometrial carcinoma, and the operation oftreatment of70patients as the experimental group, and another scheduled for uterusoperation in100patients, including58cases of atypical hyperplasia of endometriumas hyperplasia group, normal uterus endometrium of42cases as the control group.Were carried out to detect the expression in normal endometrium, atypical hyperplasiaendometrium and endometrial cancer tissue XIAP, XAF-1and Smac byimmunohistochemical method. And the relationship with XIAP, XAF-1and Smac inpatients with clinical data.Results:1. XIAP is mainly expressed in the cytoplasm, brown yellow staining. XIAPalmost no expression in the control group, the positive expression in hyperplasiagroup increased significantly (P <0.05), and with the EIN level increased, the positiveexpression rate of XIAP also increased in the experimental group significantlyincreased compared with the control group, the positive expression and proliferationgroup (P <0.05).2. XAF-1and Smac mainly expressed in the nucleus, brown yellow staining. In the control group the positive expression rate was significantly higher than the othertwo groups (P <0.05) in hyperplasia group, the positive expression rates were lowerthan that of control group (P <0.05), and increased with the level of the EIN, thepositive rate gradually decreased, the positive rates of XAF-1and Smac in theexperimental group was significantly decreased (P <0.05), almost no expression.3. Expression of XIAP, XAF-1and Smac and were independent of age (P>0.05),and compared with pathological staging, histologic grade and myometrial invasiondegree and whether there are metastasis (P <0.05). Pathological staging, histologicgrade and myometrial invasion is higher the greater the degree of positive expressionof XIAP, and the presence of distant metastases in patients with tissue sample ratehigher, the positive expression of XAF-1and Smac ratio is lower, with significantdifference (P <0.05).4. The expression of XIAP and XAF-1were negatively correlated, Smac inendometrial carcinoma (r=-0.782,-0.839, P <0.001); the expression of XAF-1andSmac were positively correlated in endometrial carcinoma (r=0.831, P <0.001)Conclusion:1. Overexpression of XIAP, low expression of XAF-1and Smac, may be thecause of the endometrium atypical hyperplasia, and lead to early formationmechanism of endometrial carcinoma is one of the.2. Expression of XIAP, XAF-1and Smac with pathological staging, histologicgrade and myometrial invasion degree and whether there are metastasis, can be usedas the initial prognosis.3. Negatively correlated with expression of XIAP in endometrial carcinoma, theexpression of Smac and XAF-1; XAF-1was positively correlated with Smac. |
