Objective:Through purifying a monoclonal antibody—Bladder Cancer Cell Monoclonal Antibody1 (BcMab1) which can specially recognize human bladder urothelial cancer (BUC) cell in patients’ urine and preliminary make prediction about its clinical diagnostic value in the early non-invasive bladder urothelial cancer (BUC).Methods:Purificating BcMabl which was obatained by the of prophase preparation, then measure its concentration using enzyme standard meter and combine it with Magnetic nanoparticles (MnPs) to harvest the immune magnetic particles BcMAbl—MnPs after BcMabl is modified.106 urine specimens were collected in the Second Affiliated Hospital of Kunming Medical University, the Third Affiliated Hospital of Kunming Medical University (Yunnan Province Tumor Hospital) from August to the end of January in 2015, all of which 32 cases of urinary system benign disease,11 cases of prostate cancer,10 cases of kidney cancer,48 cases of bladder urothelial carcinoma and 5 cases of normal specimens were included, The urine specimens were centrifuged, then the supernatants were discarded and urine sediment were collected. With the magnetic action, the BcMabl— MnPs will adsorb BUC in the urine sediment, using high-power inverted microscope to observe the cells which were adsorbed by BcMabl— MnPs after giemsa’s staining. Through that, make the judgment about the specificity and sensitivity of BcMabl in early diagnosis of bladder carcinoma and its value in clinical application.Results:The concentration of the purified bladder cancer monoclonal antibody BcMabl is 2.01 mg/mL.MnPs are combined with BcMabl after BcMabl is modified, then we could collect the BcMAbl—MnPs complexes. It is only showed strong positive reaction with bladder cancer tissue and EJ cells. Experimental results show that, in the BUC group, BcMabl can specifically capture urothelial cancer cells, and the sensitivity can reach to 80%, This is significantly higher than the rest of the other groups.Conelusion:As a new type bladder monoclonal antibody with high sensitivity and specificity, BcMabl can be used as a good indicator for the diagnosis of bladder cancer in the future. And this detection method is convenient and fast obviously, which can be considered as a new method in the early diagnosis of bladder urothelial carcinoma.
