Objective:To investigate the mechanism of drug resistance of oxaliplatin resistant cell line HCT116, analysis of the effect of changes in the level of LETM1 protein.Methods:85 cases of breast cancer tissues were collected and performed immunohistochemical detection of CD1a, CD83 expressions in TIDC of breast cancer tissues, to discuss its relationship with clinicopathological features including ER, PR, HER-2, TNM staging, diameter of tumor, transfer, neutrophils and lymphocytes ratio (NLR) of breast cancer, and further to understand the characteristics, functions, immune and anti-tumor mechanism of TIDC in breast cancer, so as to provide effective evidences for the metastasis and immune escape mechanisms of breast cancer.Results:Comparison of HCT116-OxR and HCT116 cell lines, HCT116-OxR cells found reduced mitochondrial oxygen consumption is reduced, the content of mitochondrial ATP, and the content of ATP in cytoplasm increased; (2) the comparison of HCT116-OxR and HCT116 cell lines, found that the medium glucose head sugar decreased, on the contrary the lactic acid content increased; (3) in the HCT116-OxR cell line HIF-1 a, Glu-1, HK-2 and LETM1 protein expression level increased, the inhibition of glycolysis by 3-BrPA, reversed in HCT116-OxR cells HIF-1, Glu-1, HK-2 and LETM1 alpha protein expression; (4) the inhibition of glycolysis by 3-BrPA, increased the sensitivity of HCT116-OxR to oxaliplatin.Conclusion:1 HCT116-OxR cells through the inhibition of mitochondrial function, increased glycolysis to produce ATP, leading to resistance to oxaliplatin; the content of ATP 2 in cytoplasm of colon cancer cells is the key cause of oxaliplatin resistant; resistance to chemotherapy for 3 colorectal cancer cells, probably by increasing the expression of LETM1 protein level, thus inhibition of mitochondrial function, caused by the content of ATP in cytoplasm increased.
