Atractylodes lancea (Thunb.) DC. is perennial herb in Compositae and has effects of dampness and spleen, dispelling wind and cold, and eyesight and so on. It’s a common-used clinical medicine and geo-authentic crude drugs and recognized as the good variety in Jiangsu province, especially in Maoshan area. The current study of Atractylodes lancea concentrated more on the chemical constituents and pharmacological action. However, its therapeuti material basis has not yet been elucidated, resulting in the series of implementation difficultly, such as metabolism process, mechanism and bio-transformation, which directly restricts the development of it. Therefore, studying on therapeuti material basis of Atractylodes lancea and formulating the relevant quality standards with clinical efficacy is imminent. Based on the hypothesis that only the constituents migrating to blood after oral administration of TCM have the probability to be effective, serum pharmacochemistry and serum pharmacology strategy were applied to study the effective compounds in rat plasma after oral administration of Atractylodes lancea.The medicate serum were detected by HPLC and UPLC-MS, in order to reveal the main pharmacodynamic material basis of Atractylodes lancea, provides the basis for the composition of the absorption, distribution and metabolism in vivo, to establish a new method for analyzing the effective compounds in TCM.(1) Study on the fingerprints and UPLC-MS component analysis of Atractylodes lancea decoction. Traditional mode for TCM is decoction and oral administration. Based on the records, the fingerprints of Atractylodis lanceae decoction is established by HPLC method. The research shows that there were 26 different compounds in Atractylodis lanceae decoction,23 of them were identified. The average contents of main constituents of Atractyloside A in Atractylodes lancea decoction was 2.775 mg-g-1. The study initially established quality control method of Atractylodes lancea.(2) Study on serum pharmacochemistry of Atractylodes lancea. The constituents of medicine into the blood are the effect substances in vivo. In the study, the fingerprint of medicated serum was established, by using the same analysis method in virto. Through comparing and predicating of decoction, medicated serum and blank serum, the research shows there were fifteen different compounds in the Atractylodes lancea which could be absorbed by rats, eleven of them were prototype and the other four were metabolite products. After identification, the prototype components were Atractyloside C, Wogonin, Stigmasterol, 3,5-dimethyl-4-hydroxy benzoic acid, Dandelion sterol acetate, Phylloc ladanol, β-farnesene, Atractylodin, Atractylolide IV, HinesoL Constituents absorbed into blood were syringin, β-amyrin acetate and 3,4,11-trihydroxyguai-9-ene-11-O-β-D-glucopyranoside. The eleven components absorbed into blood are probably the material basis of Atractylodes lancea used for exerting efficacy.(3) Study on serum pharmacology of Atractylodes lancea. Using serum pharmacological method, the antioxidant activity of Atractylodes lancea decoction and its medicated serum was evaluated by three methods, namely, the total reducing ability, scavenging free radical (DPPH· OH-·、O2-·) and the effect on several enzymes (GSH-Px、MDA、SOD、CAT). The H2O2-based oxidative injury model was established by cultivating rat cardiomyocytes of H9c2 in vitro.The results showed Atractylodes lancea decoction and its medicated serum have high antioxidant activity, which explain that the eleven components absorbed into blood are probably the material basis used for antioxidant activity.(4) Analysis of urine components of Atractylodes lancea decoction which could be absorbed by rats. It is necessary to study the metabolites in rat urine, which helps to clear the metabolism process and material basis in vivo. Aanalysis of the compositions of rat urine in two, six and twenty-four hours after administration, shows that component peak is more and higher in two hours, but the peak were least in twenty-four hours and two peaks area increased. The research shows that there were eleven compounds were detected in the urine and five of them performed the same as those in the blood, with the same resertative time and absorbance optical spectrum. These five compounds were considered as the same compounds as those in the blood serum.The research basically cleared the effective components into blood of Atractylodes lancea used for exerting efficacy. The serum pharmacochemistry of Atractylodes lancea needs further investigation, which may lay a foundation for the exploration of the active ingredients.
