The Effect Of Buzhong Yiqi Decoction On TGFβ-3、COL1A1、COL3A1 And Its Mechanism Of Treating POP

Objective: The article is designed to explore the function of Buzhong Yiqi Decoction for procidentia of pelvic cavity via the experiment of Molecular Cell Biology. Whether the TGFβ-3、COL1A1、COL3A1 can be the index of the valid treatment will be confirmed by the comparison of m RNA expression levels of TGFβ-3、COL1A1、COL3A1 between the patients’ pelvic organ prolapse(POP) and the main uterosacral ligaments of normal uterus. In order to find out the possible mechanism and explore the possibility treatment of Buzhong Yiqi Decoction for POP,we had done a lot of work in culturing vitro cell, dosing and detecting the m RNA expression levels of TGFβ-3、COL1A1、COL3A1 to provide the theoretical evidences.Methods:1.We selected 40 patients of transabdominal or per vaginam total hysterectomy.Among them, 20 patients with acute pelvic prolapse by western medicine diagnostic criteria and traditional medicine diagnostic criteria are selected in Research Group(RG), 20 of them fell into non-oarium functional tumor and cervical intraepithelial neoplasia are selected in Normal Control Group(NCG). During the operation, the 1cm*1cm main ligament ofvaginam and sacral ligament are selected and completely ground of liquid nitrogen to tissues of c DNA and detected the manifestations of TGF β-3 、 COL1A1 、 COL3A1 m RNA via real-time fluorescence quantitative PCR determination. In sequence, they are analyzed in statistics.2.We selected 1 patient with non-oarium functional tumor and cervical intraepithelial neoplasia who need to be treated by transabdominal total hysterectomy. On operation, the 1cm*1cm vaginam sacral ligament are gathered to culture the primary cell. When the cell is cultured to fourth generation, the Buzhong Yiqi Decoction in 20g/L、10g/L、5g/L、2.5g/L、1.25g/L、0.625g/L、0.3125g/L shall be added in culture medium respectively. Each concentration is duplicated for 3 times.On the fourth day, the total RNA of Medicine-adding Group cell shall be abstracted and the exression of TGFβ-3、COL1A1、COL3A1 m RNA detected by real-time fluorescence quantitative shall be analyzed in statistics.Results:The detection result of tissular RNA: TGFβ-3:The m RNA expression levels of TGFβ-3 of POP group is significantly higher than that of NCG; COL1A1:The m RNA expression levels of COL1A1 of sacral ligament is obviously lower than that of NCG; COL3A1:The m RNA expression levels of COL3A1 of POP group is significantly lower than that of NCG. All these differences have statistic significance(P<0.01). The m RNA expression levels of TGFβ-3、COL1A1、COL3A1 of cardinal ligament of uterus and sacral ligament have no remarkable difference, indicateing no statistic significance(P>0.05).The detection result of fibroblast RNA with Compared blank comparison group, the m RNA expression levels of TGFβ-3、COL1A1、COL3A1 increased remarkably after administration, which contributed to statistic significance(P<0.01). With theincrease of concentration, the m RNA expression levels of TGFβ-3、COL1A1、COL3A1 linearly rise.When the concentration increases to 2.5g/L, the m RNA expression levels of three factors reach the peak value. With the further of concentration, the manifestations of three factors fall down.And the effect of Buzhong Yiqi Decoction on TGFβ-3, COL1A1 and COL3A1 is similar.Conclusion:1.The m RNA expression levels of TGFβ-3、COL1A1、COL3A1 of POP and NCG exist remarkable difference, which demonstrated that these three factors are closely related to the procidentia of pelvic cavity, So we can declare that these factors may be the reliable standards of deciding whether Buzhong Yiqi Decoction is valid for POP.2. The adjustment of m RNA expression levels of TGFβ-3、COL1A1、COL3A1 may related to the treatment of POP by using Buzhong Yiqi Decoction. But the further research is still in store for specific function target and mechanism.3. A separate application to Buzhong Yi Qi Decoction has certain feasibility and the clinical experiment of large sample can test these conclusions in clinic.
