| Objectives:1.Immunohistochemistry was used to detect the expression of collagen Ⅲ,MMP-1,TIMP-1 and TGF-β1 in the anterior vaginal wall,so as to provide a theoretical basis for the pathogenesis of pelvic organ prolapse.2.Objective to analyze the relationship between the expression of collagen Ⅲ,TIMP-1 and TGF-β1 in the anterior vaginal wall of pop patients and the clinical stage of pop,and to explore the pathogenic factors,so as to provide the basis for the pathogenesis.3.Object to analyze the expression of TIMP-1,TGF-β1 and collagen Ⅲ in the anterior vaginal wall of pop patients,so as to provide the basis for the pathogenesis.Methods:From March 2020 to December 2020,28 cases of pop patients who met the inclusion criteria in the First Affiliated Hospital of Kunming Medical University were selected as pop group,According to the quantitative staging of pelvic organ prolapse(POP-Q),they were divided into grade Ⅱ in 7 cases,grade Ⅲ in 16 cases and grade Ⅳin 5 cases.and 30 cases of patients who underwent hysterectomy and/or double appendectomy without pop due to benign gynecological diseases were selected as control group.The expressions of collagen Ⅲ,MP-1,TIMP-1 and TGF-β1 in the vaginal wall were detected by immunohistochemistry,and were interpreted by four grade method.The results of interpretation were statistically analyzed,and the expression differences of collagen Ⅲ,MMP-1,TIMP-1 and TGF-β1 in vaginal wall tissue between pop group and control group were compared;the relationship between the expression of collagen Ⅲ,TIMP-1 and TGF-β1 in vaginal wall tissue of pop group and clinical stage was analyzed;the correlation between the expression of collagen Ⅲ,TIMP-1 and TGF-β1 in vaginal wall tissue of pop group was analyzed.Results:1.There was no significant difference in age,BMI,menopause and parity between pop group and control group(P<0.05).2.The IRS score of collagen Ⅲ in pop group was 1.93± 1.30,and that in control group was 4.78± 2.19.The IRS score of collagen Ⅲ in pop group was significantly lower than that in control group(P<0.05).The IRS score of MMP-1 in the vaginal wall of pop group was 4.80(3.83,6.35),while that of the control group was 3.15(1.63,6.55).There was no significant difference in the IRS score of MMP-1 between the two groups(P>0.05).The IRS score of TIMP-1 in vaginal wall tissue of pop group was 1.59±1.11,and that of control group was 3.85 ± 2.19.The IRS score of TIMP-1 in pop group was significantly lower than that of control group(P<0.05).The IRS score of TGF-β1 was 1.47 ± 0.79 in pop group and 2.64± 1.42 in control group.The IRS score of TGF-β1 in pop group was significantly lower than that in control group(P<0.05).3.According to POP-Q staging method,the IRS scores of collagen Ⅲ,TIMP-1 and TGF-β1 in vaginal wall tissue of patients with grade Ⅱ,Ⅲ and Ⅳ pop gradually decreased with the increase of degree,and the differences were statistically significant(P<0.05).4.In pop group,the expression of collagen Ⅲ was positively correlated with TIMP-1(r=0.381,P<0.05)and TGF-β1(r=0.519,P<0.05);the expression of TIMP-1 was positively correlated with TGF-β1(r=0.270,P<0.05).Conclusions:1.Collagen Ⅲ,TIMP-1 and TGF-β1 are minimal expression in the tissues of POP patients.Collagen Ⅲ,TIMP-1 and TGF-β1 may be linked to the occurrence of POP.2.The expression of Collagen Ⅲ,TIMP-1 and TGF-β1 be linked to the development of POP.3.There is a definite correlation between Collagen Ⅲ,TIMP-1,TGF-β1 in the anterior vaginal wall tissue of POP patients.Collagen Ⅲ is positively correlated with TIMP-1 and TGF-β1,and the expression of TGF-β1 is positively correlated with TIMP-1.There may be a certain synergy between the three,and jointly participate in the occurrence and progress of the disease. |
