This topic set out from the Chinese medicine Qi theory of its own context that developed by itself.has carried on the research and induction of the theory of traditional.Combining the previous research,take senility as the break-through point to explore the relationship between Qi and senility and Study on Anti-aging Mechanism of Dabuyuanjian. With Caenor-habditis elegans as an animal model,To investigate effect of Dabuyuanjian prescription On anti-senility of Caenorhabditis elegans. Aimed at providing a theoretical basis and experimental evidence for further more depth research from theory of TCM study and vitality injury pathogenesis and elderly population strength loss research,the development research of the product and the system of health care in peiyuan solid methods,also about the mechanism of experimental study and anti-aging therapies and application of prevention and treatment of geriatric on the basis of the peiyuan solid. Method: 1、Theoretical discussion:Collate the ancient and modern literature about the Qi of Chinese medicine,summarized the basic concepts of yuanQi,the theoretical connotation of Medicine. Study of the formation and development, and the replenishing of yuan Qi,the development and impact. To investigate the correlation between yuan Qi and the senility,explain the relationship between the yuan Qi damage and senility pathogenesis,replenish yuanQi to prevent senility.This paper considers the Qi theory of TCM has a distinctive strength characteristics of Chinese, which has accumulated valuable experience for human delay aging,prevention and treatment of age-related diseases. Through repeated explorations to found the effect of yuan Qi. 2、Experimental study:Using Caenorhabditis elegans model of senility, to study the effect and mechanism of the traditional Chinese medicine Da Bu Yuan Jian which was used for Reinforcing primordial qi for senility. By life experiments to observe Dabuyuanjian’s effect on c.elegans lifespan; Through fertility test, we can observe effect of c.elegans’ fertility; by acute heat stress and oxidative stress experiments to observe the change of life under the stress station.Methods RT-PCR semiquantitative technique was applied to show the relationship between Da Bu Yuan Jian and ageing. Results:Da Bu Yuan Jian can significantly extend the average life and maximum life span of c.elegans(p < 0.05, p < 0.01), The survival curve obviously moves to the right.and no damage in fertility; Da Bu Yuan Jian can significantly extend the life of the c.elegans in oxidative stress and thermal stress environment(p < 0.05, p < 0.01). Methods RT-PCR semiquantitative technique was applied to gene tests.The Caenorhabditis elegans which has been oxidative damage and thermal stress environment were treated in medium of containing Da Bu Yuan Jian under normal conditions for some time. The results showed that the expression of Daf-16 、 Hsp-16.2 and SOD-3 was increased in Caenorhabditis elegans(P<0.05,P<0.01). Especially the Hsp-16.2 gene expression increased significantly(P<0.01). Da Bu Yuan Jian s ability of enhancing c.elegans’ stress resistance probaly is the mechanism. Conclusion: 1.Enough vitality is an important principle of anti-aging.2.Da Bu Yuan Jian can delaying the aging of c.elegans. 3.Da Bu Yuan Jian s ability of enhancing c.elegans’ stress resistance probaly is the mechanism.
