| Aging is characterized by a progress that loss of physiological integrity,leading to impaired function and increased vulnerability to death with the increase of age.Aging has been proposed as the main risk factor for these prevalent diseases,such as cardiovascular,cancer,neurodegenerative disorders,diabetes,stroke,and infectious diseases.Although aging is inevitable,it can delay the aging process.Therefore,there is a hot topic for Screening of Anti-aging Compound through Anti-aging mechanisms.Anti-aging compound can delay aging,improve the quality of life and improve the efficiency of life,reduce pathological death,make life closer to the physiological limits.Anti-ageing compounds can be divided into natural compounds and synthetic compounds,Among them,Astragaloside IV,Ginkgolide,Galangin,Berberine and Rosmarinic Acid are all effective components extracted from natural plants.Studies have shown that Anti-aging compounds have a variety of pharmacological effects including anti-oxidative,anti-inflammation,regulation nervous system to improve neurodegenerative diseases and so on.Caenorhabditis elegans is an excellent model for identifying genetic and pharmacologic interventions in lifespan assay,due to the aging signaling pathways were conservative from yeast and worms to mammals.Our research use C.elegans to screen anti-aging compounds of AS-IV,GA,GB,RA,Galangin and Berberine.Results show that:1.Worms raised at 20℃ on NGM plates containing different concentration of AS-IV could slightly extend lifespan by 4.5%~12.0%.The different concentration of GA and RA could slightly extend lifespan by 5.2%~11.6%and 2.4%~8.8%.2.When the maximum life extension rate is more than 10%,it indicates that the compound has significant anti-aging activity(P<0.01).100 μM of AS-IV has the most significant effect of displaying the largest lifespan by 12.12%,1000 μM of GA and RA displays the largest lifespan by 11.6%and 8.8%;while 200 μM of Galangin has the most significant effect of displaying the largest lifespan by 8.79%,and 1000 μM of GB has the most significant effect of displaying the largest lifespan by 6.88%,and 500 p.M of Berberine has the most significant effect of displaying the largest lifespan by 4.88%.Therefore,AS-IV,GA and RA have significant anti-aging activities,while Galangin,GB B and Berberine have no significant anti-aging activities.3.Worms raised at 35℃ on NGM plates shown that 100 μM of AS-IV showed a significantly high temperature resistance,increasing the lifespan rate up to 25.14%,however,1000 μM of GA and RA displays the lifespan by 23.04%and 21.23%.The most significant effect is AS-IV,as the same as lifespan.Taken together,we can draw a conclusion that:(1)AS-IV,GA,RA can increase the normal lifespan of C.elegans,improves the thermos-tolerance,It has certain anti-aging activity.However,Galangin,GB and Berberine had no significant anti-aging activity.(2)AS-IV increased the number of nematode eggs and decreased the fat content,which may be related to the reproductive signaling pathway.And daf-16::GFP drugs were obviously nucleated,and the anti-aging mechanism may be related to daf-16. |
