Objective:In vitro observation and evaluation of different therapeutic principle under the guidance of traditional Chinese medicine to TGF-β1 inducing embryonic lung fibroblasts proliferation,apoptosis and cell factor synthesis and secretion.Methods:Experiment with the embryonic lung fibroblasts as the research object,and set up blank control group,model group,ginkgo biloba extract low,medium and high concentration groups,Shenmai injection of low,medium and high concentration group,matrine injection low,medium and high concentration group,after completion of training,cells,respectively each COL-Ⅰ,COL-Ⅲ,α-SMA,ECSOD,PDGF protein and mRNA in the OD value of cell proliferation and apoptosis rate.Results:Model group cell proliferation OD value higher than the control group obviously, suggests that TGF-β1 can obviously promote the lung fibroblast proliferation;Cell proliferation OD value of each drug group were significantly lower in the model group,the cell growth inhibition rate and shenmai injection group was obviously higher than that of other drug group,and high concentration significantly higher than the middle and low concentration group,after statistics processing,there was a significant difference(P<0.05).The experiment show that shenmai injection on lung fibroblasts proliferation inhibition of the most obvious and has obvious correlation with drug concentration.Model group significantly lower than the control group,cell apoptosis rate showed that TGF-β1 can obviously inhibit lung fibroblast apoptosis;Apoptosis rate of each drug group were significantly higher in the model group, the shenmai injection group is significantly higher than other drugs,and high concentration significantly higher than the middle and low concentration group,after statistics processing,there was a significant difference(P<0.05).The experiment show that shenmai injection on lung fibroblast apoptosis role in promoting the most obvious and has obvious correlation with drug concentration.Model group cells COL-Ⅲ,COL-Ⅰ,α-SMA content than the control group were significantly higher, TGF-β1 can obviously promote the lung fibroblasts COL- Ⅲ,COL-Ⅰ,α-SMA synthesis and secretion;Each drug group of cells COL-Ⅲ,COL-Ⅰ,α-SMA protein content were significantly lower in the model group,among which COL- Ⅲmatrine,COL-Ⅰ,α-SMA protein content was lower than that in group other drugs, and high concentration significantly lower than the medium and low concentration group.The experiment show that matrine injection by inhibiting the COL-Ⅲ,COL-Ⅰ,the synthesis and secretion of α-SMA play a role of anti fibrosis.Cell ECSOD content than control group in model group were significantly lower,show that TGF-β1 can obviously inhibit lung fibroblasts ECSOD synthesis and secretion;Drug groups each cell ECSOD protein content were significantly higher in the model group,in which the protein content of ginkgo biloba extract group ECSOD significantly higher significantly better than other drugs,and high concentration group,the low concentration group;Model group cell PDGF content than the control group were significantly higher,shows that TGF-β1 can obviously promote the synthesis and secretion of lung fibroblasts PDGF;Each cell PDGF protein drug group was obviously lower than other drugs group,PDGF protein content of ginkgo biloba extract group was obviously lower than that of other drugs,and high concentration group significantly lower than the medium and low concentration group. The experiment show that ginkgo biloba extract by promoting fibroblasts ECSOD,inhibit the synthesis and secretion of of PDGF play a role of anti fibrosis.Conclusion: Ginkgo biloba extract,shenmai injection and matrine injection can affect lung fibroblasts proliferation,apoptosis,and the production of a related cytokines secreted rise to slow down and the role of the occurrence and development of the pulmonary fibrosis. Ginkgo biloba extract mainly by influencing the fibroblasts ECSOD,PDGF synthesis and secretion of cytokines,such as to infer the main method for activating blood circulation through the body oxidative stress state of antagonism,improve alveolitis,play a role in the treatment of pulmonary fibrosis;Shenmai injection on lung fibroblasts proliferation inhibition and apoptosis of the most significant,to infer,qi mainly by inhibiting lung fibroblasts proliferation, promote apoptosis,delay the fibrosis process;Matrine injection on lung fibroblasts COL-Ⅰ,COL-Ⅲ,the influence of the alpha SMA was the most obvious,to infer,heat-clearing and detoxifying method mainly affect the synthesis of collagen,to reduce fiber deposition of extracellular matrix,thereby reduce the degree of fibrosis.
