Cortex Phellodendri is the dry bark of Rutaceae plants Huang Shu pi or Cortex Phellodendri. Huang Shu pi is also called "Cortex Phellodendri Chinensis," and Cortex Phellodendri is namely "Cortex Phellodendri Amurensis". Which both have the character of bitter and cold and effects of clearing heat and drying dampness, removing firelight and excepting steam, curing poison and sore. The results have showed that Cortex Phellodendri has extensive pharmacological activities, such as antibacterial, antiviral, relieving cough, depressurization and enhance immunity, etc. As a traditional Chinese medicine, Cortex Phellodendri display high clinical application value and development potential. Alkaloids is the main active ingredients in Cortex Phellodendri, especially berberine, the content of which reaches to1.4%-5.8%. In addition, Jateorrhizine, Phellodendrine, Palmatine, Obaculactone and Obakunone also occupy certain proportion. This article mainly study on the extraction process of the main alkaloids and obaculactone in Cortex Phellodendri.In order to optimize the raw material source, we detected and compared the content of berberine of different ages, different seasons and different parts of the Cortex phellodendri by HPLC. The results showed five-years growing Cortex phellodendron bark harvested in November has the highest content of berberine and determined as the best material, which may played a guiding role for the industrial production of the enterprise.The experiment established an percolation extraction method for phellodendrine and obaculactone from Cortex Phellodendri. The factors of crush granularity, extraction temperature, percolate pH, solid-liquid ratio and percolate cycle were taken in account. The result showed the optimum extraction conditions for phellodendrine and obaculactone were: the crush granularity is0.1cm, the ratio of solid-liquid is1:20, extraction temperature is90℃, percolate cycle is10times, and percolate pH is Under these conditions, the extraction efficiency could be improved.Taking salting-out method and ion exchange process to refine the total alkaliod, and compared metastasis rate of two methods. In the salting-out method, the parameter of minera salt proportion and salting-out concentration were investigated, the optimal salting-out concentration was12%NaCl solution. In the ion exchange process, we mainly investigated the concentration of ammonia in the absorption and desorption process, the results showed that10%ammonia was the optimal eluting reagent. Finally, the metastasis rate of two methods were measured by HPLC and the effection of salting-out for alkaliod refining was better than the methord of ion exchange.
