| Objective: Combining Multivariate Statistical Analysis to Evaluate the Quality of Effective Matter of Cortex Moutan from Different Areas.Meanwhile,the pharmacodynamic material basis of cortex moutan was investigated by means of theory and method of serum pharmacochemistry.Methods:Using orthogonal design method to optimize the best ethanol reflux extraction process of Cortex Moutan;Ultra high performance liquid chromatography(UPLC)method is adopted to establish the different origin of Cortex Moutan fingerprint and method for the determination of component content;Determination of trace elements in Cortex Moutan by inductively coupled plasma mass spectrometry;The quality of Cortex Moutan in different habitats was compared and analyzed by multivariate statistical analysis,such as principal component analysis,cluster analysis and discriminant analysis.By using the optimized conditions of chromatography and mass spectrometry,UPLC-Q-TOF-MS was used to analyze and identify the components in and out of the body.Results: The optimum extraction process of ethanol reflux was 90% of ethanol volume fraction,and 60 min was extracted at 1:10,and 2 times were extracted.Through the study of fingerprint patterns,15 common peaks were identified and eight peaks were identified.There was a good aggregation effect in each area,and the possible chemical components of the differences were selected.A method for the determination of 8 main active ingredients of Cortex Moutan,including gallic acid,oxidized paeoniflorin,catechin,paeoniflorin,benzoic acid,benzoyl oxidized paeoniflorin,benzoyl paeoniflorin and paeonol was established.Combined with ICP-MS analysis of Cortex Moutan medicinal materials quality and the relationship between the trace elements,it is found that the original region medicinal materials of Cu、Fe、 Co、Zn、Ti content is higher,the principal component analysis to select the six main factors,it is concluded that the characteristics of cortex moutan inorganic elements Cu、Fe、Ti、Sn、Mo、Co、Hg、Zn、Cr.43 chemical components were identified by UPLC-Q-TOF-MS and UNIFI data processing system,and 11 kinds of blood components were screened.Conclusion: As a complex system,Chinese medicine can reflect its own characteristics more effectively.The purpose of this study is to comprehensively evaluate the quality of Cortex Moutan by means of fingerprints,multi-component contents and trace elements.Meanwhile,we guided the serum pharmacochemistry as a guide to screen the components of blood from Cortex Moutan,and lay the foundation for the research of its effective substance foundation. |
