Research On The Regulation Of BMSCs’ β-galactosidase Activity With Bushen Huayu Shengxin Decoction

Objective : To investigate the regulation of Bone Mesenchymal Stem Cells’ β-galactosidase activity with Bushen Huayu Shengxin Decoction and the mechanism about how the decoction delay the cells’ senility. Method: 40 rats were divided into Bushen Huayu Shengxin Decoction group and normal saline group random Ly.The rats were intragastric administration for 7 days,and drug serum was prepared through picking blood from abdominal aorta 2 hours after the first administration at the 7th day.The 100 newborn rats were proceed the extraction, incubation and isolation purification of BMSCs;4g/L D-galactose was used to establish the aging model of BMSCs. The natural aging cell and BMSCs of aging model were divided into Bushen Huayu Shengxin Decoction group and normal saline group random Ly,then drug serum was used to culture those cells. Light microscopic was used to observe the BMSCs’ morphologic change of every generation. MTT assay was used to test the BMSCs activity change of different concentration of drug serum.Colorimetric method was used to detect the β-galactosidase activity of BMSCs of all the groups. Results:Light microscopic show that the normal BMSCs shape are same.The major shapes of cells are spindle,triangle,and the BMSCs fusion together like vortexes.The BMSCs shapes of aging model group are flat triangle,oval,and the aging cells grow slowly.The result of MTT show that BMSCs grow slowly at 1st and 2nd day,this illustrates that the 1st and 2nd day is incubation period of proliferation.At 3rd day the cells proliferate fast.The BMSCs survival rate of Bushen Huayu Shengxin Decoction group are higher than normal saline group,DMEM control group;The best concentration of drug serum is 10%.With the generation gaining,the β-galactosidase activity of BMSCs in each group of aging model group and normal group is increasing.The activity of Bushen Huayu Shengxin Decoction group is lower than that of normal saline group,has the statistical significance(P<0.05). Conclusion:Under the condition of natural aging and induced aging,Bushen Huayu Shengxin Decoction can decrease the β-galactosidase activity of BMSCs,and the decoction can delay the senility of BMSCs.
