| Objectives Study the endometriosis trends and characteristics of the disease in recent years,to screen out the risk factors and protective factors of endometriosis pathogenesis. And from the angle of epigenetics, study the relevance of IFN-γ gene promoter methylation and endometriosis and its risk factors. The purpose lies in enhancing the awareness of the disease among medical workers, and providing scientific and feasible prevention measures for the prevention of endometriosis, providing new scientific basis for the assessment of endometriosis risk, prediction, molecular diagnosis and treatment.Methods Select 800 cases gynecological inpatients who were diagnosed as endometriosis patients as cases from January, 2009 to December, 2014, in Tangshan workers hospital.Random select 800 patients who lived in gynecological ward at the same time,beared annex block or abdominal pain underwent open operation or laparoscopy package and except endometriosis of the others as control group. Collect the medical records of the clinical datas of the two groups and analyze retrospectively, and conduct cases in casecontrol studies. Select 45 patients who beared ovarian endometriosis ectopic endometrium as case group, from June, 2013 to June, 2014 in our hospital. Select 45 cases for a variety of benign diseases undergoing total hysterectomy and postoperative pathology confirmed patients with normal endometrial uterine endometrium as control group. Using MSP and RT-PCR method for semi quantitative detection of the expression and methylation status of IFN-γ in ectopic endometrium and normal endometrium,to study the relevance of IFN-γpromoter methylation and endometriosis and its risk factors.Results 1 The peak age of endometriosis patients among 25-48 years old.The endometriosis patients accounted for the proportion of gynecological patients increased year by year.Ovarian endometriosis accounted for a larger proportion, and there is no significant difference in the occurred side,and the majority of diameter is 5-6cm. The level of CA125 is higher than other categories of endometriosis, and the main operation mode is laparoscopic operation. 2 Blood type A, dysmenorrhea, abortion for many times, caesarean operation history, placed IUD, a long menstrual period, and heavy menstrual volume are risk factors of endometriosis.Give birth to more children and long menstrual cycle are protective factors of endometriosis. The age, nation, marital status, place of birth, history of past illness, other gynecological operation history, uterine position of patients with endometriosis,compared with the control group, have no statistical significance(P>0.05).3 The comparison of cases and control patients with clinical typical signs and symptoms are pelvic pain, poor activity, tenderness and elevated levels of CA125(P < 0.05). 4Interferon gamma gene promoter region activation occurs aberrantly methylated in ectopic endometrial tissue, and leads to low expression of IFN-γ m RNA. 5 There is correlation among heavy menstrual volume,dysmenorrhea,placed IUD and IFN-γ promoter methylation(P>0.05), and other risk factors are found no correlation(P>0.05). 6 Gamma interferon promoter methylation level and CA125 level have no correlation in ectopic endometrial tissue(Z=-1.645, P>0.05), and the expression of IFN-γ m RNA have no correlation with CA125 level(r=0.263, P>0.05).Conclusions 1 Blood type A, dysmenorrhea, abortion for many times, caesarean operation history, placed IUD, a long menstrual period, and heavy menstrual volume are risk factors of endometriosis.
