Objectives To study the changes of Hif-1α and VEGF expression after cerebral ischemia in rats and its mechanism.Methods Sixty adult male SD rats were divided randomly into 2 groups: the experimental group and the control group, 30 rats in each group. After the success of modeling 2h, the experimental group were injected with HIF-1 plasmid PBS buffer solution, the control group was injected the same volume, according to the different time points after injection of 6h, 24 h, 48 h, 72 h, 7d five time points,6 rats in each subgroup. TTC staining method was used to observe the cerebral infarction area of rats in each group match to determine the degree of staining, tissue ischemia brain; using immunoblot assay to detect HIF-1 alpha and VEGF in infarcted brain tissue protein expression.Results 1 Observe the rats cerebral infarction compared with the control group match area by TTC staining, no significant difference between the experimental group 6h, 24 h, 48 h time point of the infarct area(P>0.05), and the difference in 72 h, 7d time point, the more significant difference(P<0.01). 2 Effects of injection of HIF-1 plasmid solution on rat mortality of rats in ischemia reperfusionafter injection of HIF-1 plasmid in liquid, 4h ~ 60 h was the highest, the survival time was more than 72 h Low mortality(P < 0.01). 3 Expression of HIF-1 and VEGF test for the detection of brain infarction area in Western blottwo factor protein results in the control group and the experimental group of two kinds ofprotein expression gradually increased in 6h, 24h(P<0.01), 24 h expression was the highest, 48 h, 72 h, 7d decreased, 7d expression was the lowest(P<0.01). Compared with the control group, the expression of the experimental group at each time point of HIF-1 alpha and VEGF protein were increased, with statistical significance(P<0.05). Correlation analysis between the expression level of HIF-1 alpha and VEGF two protein expression of VEGF and HIF-1. The results show that there is a positive relationship between alpha.Conclusions 1 The plasmid HIF-1 in cerebral ischemia reperfusion injury in rats, contribute to the improvement of cerebral ischemia and infarction area, shrink.2 In cerebral ischemia reperfusion injury, HIF-1 plays a very important role in alpha, VEGF factor in this regulatory process.. |
