Objective: To observe the impacts of Rheum tanguticum on endothelial nitric oxide synthase(e NOS), endothelin(ET)and 6-keto-prostaglandin F1alpha(6-keto-PGF1α) in uric acid nephropathy rat model.Methods: Uric acid nephropathy rats model were treated with different extractions of Rheum tanguticum; Biochemical analyzer was used to measure the level of serum blood urea nitrogen(BUN),creatinine(Cr) and uric acid(UA) after three weeks; The method of ELISA was used to measure the expression of e NOS,ET,6-keto-PGF1α, and the light microscope was used to observe the pathological changes of rat’s renal tissue.Results: Compared with the normal control group,the levels of serum BUN, Cr, UA and renal index were significantly higher in model group(P<0.05); The concentration of e NOS in model group were significantly lower than the other groups(P<0.05); The concentration of ET in model group were higher than that in normal control group, rhubarb water extract treatment group and allopurinol treatment group(P<0.05), and there was no obvious difference with rhubarb ethanol extract treatment group; The concentration of 6-Keto-PGF1α was no significant difference among five groups(P>0.05); Under the microscope, there were a lot of inflammatory cell infiltration and uratic crystallization in renal tissue of model group rats;The damage of rats renal tissue in all drug groups was eliminated than that in the model group.Conclusion: Rheum tanguticum can reduce uratic crystallization inflammatory cell infiltration and the serum concentration of ET, and increase the serum concentration of e NOS. Thus it can alliviate the damage of renal endothelial cells and protect renal functions; ET decreasing effect of the Rheum tanguticum water extraction was better than ethanol extraction’s. |
