Objective To study the relationship between the parameters of Mutiple-Voxel Spectroscopy(1H-MVS)(Cho/Cr) and tumor stem cell markers such as CD133、Nestin in glioblastoma.Methods The conventional MR imaging and 1H-MVS examination were performed in 5 cases with gliomas diagnosed by the postoperative pathological examination from April 2013 to Auguest 2013. Area of interest include choline / creatine(Cho / Cr) significantly different areas, then cultured primary gliomas cells.We detect the CD133 and Nestin expression in gliomas via measuring protein level(Western Blot), morphology level(immunohistochemistry) in vivo. Then analysis the relationship between 1H-MVS parameter(Cho / Cr) and cancer stem cell marker(CD133).Take U87 glioma becoming-ball cell as glioma stem cell model in vitro.We measured choline content of glioma stem cell model and U87 glioma cell to investigate whether choline content is associated with glioma stem cell or not.Results primary culture of glioma showed that glioma exist glioma becoming-ball stem cell which express CD133.We observe that regional tumor tissues which has significant differences in the parameters of Mutiple-Voxel Spectroscopy(1H-MVS)(Cho/Cr), and which has corresponding different expression of CD133 and Nestin. The areas with high Cho/Cr ratio has higher obviously expression of CD133 and Nestin level than that with low Cho / Cr ratio, statistically significant difference(P < 0.05).After correlation analysis, parameter of H-MVS Cho / Cr and cancer stemcell marker CD133 has a positive correlation(r = 0.927, P <0.01), as well as marker Nestin(r = 0.964, P <0.01). In vitro experiment, U87 glioma becoming-ball cell with high expression of CD133 contain more choline than that adherent cultivation U87 tumor cells contain, statistically significant difference(P < 0.05).Conclusion The parameters of Mutiple-Voxel Spectroscopy(1H-MVS)(Cho/Cr) has a positive correlation with cancer stem cell marker CD133 and Nestin. It can effectively reflect the different distribution of cancer stem cell with positive CD133,Nestin in single tumors and can effectively find the concentration areas of cancer stem cell with positive CD133,Nestin to guide clinical operation.
