Objective: This paper uses a chemical drug streptozotocin(Streptozotoein, STZ)induced diabetic retinopathyrats, systematic observation of HPS(Hedysarum Polybotys saccharide,HPS) on vascular endothelial cells growth factors(Vascular endothelial grow thfactor, VEGF), the impact of pigment epithelium-derived factor(Pigment epithlium-derived facto, PEDF) and serum C- reactive protein(C-reaetive Protein, CRP) levels, the impact of the retinal histopathology,thus HPS clarify the treatment of DR mechanism of action and therapeutic targets.Methods: 80 SPF Wistar rats were randomly divided into six groups according to body weight and male:control group,model group,Duobeisi group, HPS low dose group,HPS middle-dose group,HPS high-dose group,The control group 10,and the remaining14 per group.All blank rats by intraperitoneal injection of citrate buffer line,the rats in other groups were treated with intraperitoneal injection of streptozotocin(streptozotocin,STZ,30 mg.kg-1,continuous administration of 3 days)was induced by intraperitoneal injection of STZ rats model,after the successful preparation of animal models,HPS treatment groups given hedysarum polybotys saccharide the appropriate concentration of HPS mice,the model group were given Duobeisi gavage,the control group of rats to the equivalent volume of distilled water once daily for 8 weeks. After the end of the last avage, rats were anesthetized before the femoral artery blood serum CRP content detection.And stripping the retina, retinal p-athological examination,SP immunohistochemical staining using retinal tissue of rats and expression of VEGF, PEDF.Results: 1.Each group in the rat retina pathological observation results:compared with the blank group, model group rats retinal each layer are different degree of thinning, and each layer is reduced Numbers of cells,cell arrangement disorder obviously.Compared with model group,HPS each dose groups all have different degrees of retinal thickening.And beth groups more contrast,HPS retinal thickening degree high dose group, the most obvious cell basic neatly arranged.Compared with HPS high dose,dosage,HPS low doses of HPS. Each group in the rat retina of VEGF and PEDF test results:compared with blank group, model group of VEGF expression in the retina tissue volume increases, thecontent of PEDF fell,the difference was statistically significant(P <0.05,P<0.01);Compared with model group,HPS in each treatment group and VEGF expression more be the group were lower,the expression of PEDF were higher,the difference was statistically significant(P<0.05), compared with more bass group,HPS high-dose group reduced ex-pression of VEGF and PEDF expression increased,with statistical significance(P<0.05),t-he HPS dose group,the HPS low-dose group reduced expression of VEGF and PEDF expression l have significant difference(P<0.05,P<0.01);Compared with HPS high dose group,low dose group,the HPS in HPS dose groups was statistically significant(P<0.05),beth groups more reduced expression of VEGF and PEDF expression increased with significant difference(P<0.05)Conclusion:1.Hedysarum Polybotys saccharide can reduce the blood sugar of DM rats, and can improve the damage degree of DM rats retinopathy;2. Hedysarum Polybotys saccharide can significantly inhibit the DM rat serum levels of CRP in the improvement of the DM rat inflammatory state,to protect the DMrat retina tissue have a positive effect;3. Hedysarum Polybotys saccharide allows expression of VEGF in retinal tissue of DM rats decreased the expression of PEDF rises, and play its role in protecting t-he retina and thus deter DM rats with retinal neovascularization produce, thereby preventing and delaying the development of DR.
