Objective: To investigate the expression of FAK(focal adhesion kinase), NF-κBp65(nuclear factor-kappaBP65) and VEGF-C(vascular endothelial growth factor-C) in non-small cell lung cancer(NSCLC) in order to detect their relationship between the clinical pathological feature,diagnoses and prognosis. Methods: Patients were treated by operation in The 401 st Hospital of the PLA between between December 2012 and December 2013, of them 75 patients with NSCLC were chosen as cases(specimen requirements as following: 1.those who never received the chemotherapy and radiotherapy before the surgery; 2.those who without primary tumors in other organs; 3.those whose tissues are paraffin-embedded fully). 30 adjacent lung tissues were selected randomly as controls.The histopathological classification and grading were determined by the pathologists from 401 th hospital.Immunohistochemical Elivision method was used to detect the expression of FAK、NF-κBp65 and VEGF-C in 75 NSCLC tissues and 30 adjacent normal lung tissues. SPSS 17.0 statistics software was applied for statistical analysis, and χ2 test was used for the expression of FAK、NF-κBp65 and VEGF-C in NSCLC with its clinical pathological feature, grading and comparison between the experimental group and the control group.Spearman correlation analysis was used for the relationship between FAK、NF-κBp65 and VEGF. P<0.05 was considered to be statistically significant. Results:(1)FAK and VEGF-C showed brown-yellow solids precipitation in cytoplasm was positive and NF-κBp65 showed yellow solids precipitation in cytoplasm and nucleus was positive.(2)The positive expression rates of FAK, NF-κBp65 and VEGF-C were 57.3%(43/75), 85.3%(64/75) and 65.3%(49/75) in NSCLC tissues but those were 13.3%(4/30),16.7(5/30) and 0%(0/30) in normal lung tissues(P<0.05).(3)The expression of FAK, NF-κBp65 and VEGF-C were all related to differentiation and TNM stage, in poorly differentiated lung cancer tissue was significantly higher than that in well-moderately differentiated(P<0.05). Their expression in III~IV was significantly higher than that in I~II(P<0.05).(4)The positive expression of FAK and NF-κBp65, NF-κBp65 and VEGF-C in NSCLC were related respectively(P<0.05). Conclusions:(1)The expression of FAK,NF-κBp65 and VEGF-C in NSCLC tissue were significantly higher than that in normal lung tissues, suggesting that the two may be involved in the occurrence of lung cancer.(2)The expression of FAK,NF-κBp65 and VEGF-C in NSCLC tissue were associated with tumor differentiation and tumor TNM stage. The higher degree of malignancy of lung cancer, the higher expression level of FAK, NF-κBp65 and VEGF-C, which suggests that the higher expression the poorer prognosis.(3)The expression of FAK and NF-κBp65, NF-κBp65 and VEGF-C in lung cancer tissue was positive correlated respectively. However, the interaction mechanism of them in the process of lung cancer remains to be further studied.
