| Objective: To explore the SUMO-specific proteases 1 whether have the power to influence the migration/proliferation ability in cholangiocarcinoma cell lines; To investigate the possible mechanisms underlines the influence activity; To investigate the SENP1 peotein expression of the cholangiocarcinoma tissues.Methods: First, through over-expressing SENP1(SUMO-specific proteases 1) then observed cholangiocarcinoma cell’s phenotype. Second,through transfected sh-SENP1 plasmid into cholangiocarcinoma cell lines to observe the phenotype. Third,using the MTT and transwell assay to explore the migration/proliferation ability change of the cholangiocarcinoma cells. Forth, fluorescence quantitative PCR and Western blot assays were conducted to measure the m RNA and protein expression of target proteins/genes. Fifth,extracted the total proteins of the cholangiocarcinoma tissues,then explore the SENP1 peotein expression through Western blot assays.Result: First,the overpression of the SENP1 can increase migration and proliferation ability of the cholangiocarcinoma cells. Second,When overexpression SENP1,we detected the increase of AIB1 in m RNA and protein levels. Third, a decrease of the migration and proliferation ability of the cholangiocarcinoma cells can be detected when wo knock down the SENP1 in cholangiocarcinoma cell lines. Forth, we found that the high expression of SENP1 protein in cholangiocarcinoma tissues.Conglusion: First, the change of SENP1 expression in cholangiocarcinoma cells can cause change of the proliferationã€migration and invasion ability.Second, SENP1 may activate AKT pathway through AIB1. |
