Objective: To investigate the effect and mechanism of total flavones of Chrysanthemum indicum combined with cisplatin on human osteosarcoma cell line MG-63.METHODS: MG-63 cells were treated with total flavones of Chrysanthemum indicum alone or combined with cisplatin;the cellular morphologic changes were observed under inverted phase contrast microscope; the effects of proliferation inhibition were assayed by CCK-8;the drug-drug interactions were judged through Q value calculated by King’s Formula;the apoptosis ratios of cells were analysised by flow cytometry;the expression of Bcl-2、Caspase-3 and p21 gene was detected by RT-PCR.RESULTS: Total flavones of Chrysanthemum indicum alone or combined with cisplatin could both inhibit the proliferation of MG-63 cells and induce the apoptosis of MG-63 cells,the expression of Bcl-2 was reduced,and the expression of Caspase-3and p21 was enhanced;the combination of total flavones of Chrysanthemum indicum and cisplatin revealed a synergistic effect at low concentrations, and revealed a superimposed effect at high concentrations.CONCLUSION: Total flavones of Chrysanthemum indicum combined with cisplatin can both inhibit the proliferation of MG-63 cells and induce the apoptosis of MG-63 cells,and have a synergistic effect within a certain concentration range,the mechanism of which may be related with reducing Bcl-2 and enhancing Caspase-3and p21.
