| Objective:The honeysuckle stem sugar free granules effect on acute gouty arthritis(GA)rat mo del validation for its efficacy; Through the detection of gouty arthritis model rats serum u ric acid value and effect of IL-1β in articular cartilage in synovial fluid levels of TNF-α.Study on the mechanism of Caulis Lonicerae sugar free granule in treating gouty arthritis and its effect on acute gouty arthritis. By studying the honeysuckle sugar granules on g outy arthritis in rats pathological changes, further validate its anti-inflammatory effect.Honeysuckle sugar granules prepared using the method to expand the original parties on the basis of Gout range of applications, but also laid the foundation for the further development of new drugs.Method:The 48 SD rats weighing 200±20g, half male and half female, were divided into 6groups, divided into blank control group(8 rats), model group(8 rats), colchicine grou p(8 rats), honeysuckle stem sugar free granules in high dose group(8 rats), honeysuckle stem sugarless granules in the dose group(8 rats), honeysuckle stem sugar free granule low dose group(8 rats). The data for statistical analysis.Results:① honeysuckle stem compound and western medicine can reduce the blood uric acid values of model rats compared with model group had significant difference(P<0.01), honeysuckle stem the high dose group, honeysuckle stem low dose group compar ed with western medicine group, had significant difference(P<0.05). Honeysuckle ste m middle dose group compared with the western medicine group, there was no statisti cally significant difference(P>0.05).② building 72 h after the honeysuckle stem comp ound with western medicine can make the model rats joint swelling degree decline, th ere are significant difference statistically significant(P<0.01), honeysuckle stem compo und group compared with the western medicine group, with significant difference statis tically significant(P<0.01), honeysuckle stem middle dose group and honeysuckle stem in high dose group, honeysuckle stem low dose group, with significant difference(P<0.01). ③ honeysuckle stem compound and western medicine can relieve inflammatio model in rats, 48 h after the treatment, honeysuckle stem compound compared group,the western medicine group and model group, with significant difference statistically si gnificant(P<0.01); 48 h after the treatment, honeysuckle stem middle dose group com pared with the western medicine group, with significant difference statistically significa nt(P<0.05); After 72 h, honeysuckle stem high dose group, middle dose group, low d ose group, compared with western medicine group, there was no significant statistical significance(P>0.05), honeysuckle stem high dose group, middle dose group, low dose group, comparison between way, there was no significant statistical significance(P>0.05). honeysuckle stem compound and western medicine can relieve model rats joint pa in, 24 h, 48 h after the treatment in honeysuckle stem middle dose group, western m edicine group with significant difference compared with model group statistical signific ance(P<0.01), 24 h dosing honeysuckle stem high, medium and low group there was no statistically significant difference compared with the western medicine group(P>0.05).⑤ ELISA method to detect shows: honeysuckle stem compound and western medi cine can reduce the expression of IL-1β in joint fluid, compared with model group, with significant difference statistically significant(P<0.01), honeysuckle stem high dose group, middle dose group, low dose group, compared with the western medicine group,with significant difference statistically significant(P<0.01). Honeysuckle stem in midd dle dose group compared with honeysuckle stem low-dose group, with significant diffe rence statistically significant(P<0.01); Honeysuckle stem in high dose group, honeysuc kle stem middle dose group comparison between the two groups have no significant st atistical significance(P>0.05); Honeysuckle stem compound and western medicine can reduce joint fluid model rats with the expression of TNF-α, had significant difference compared with model group, the statistical significance(P<0.01), honeysuckle stem mi ddle dose group compared with blank group, there was no statistically significant diffe rence(P>0.05), honeysuckle stem compound group compared with the western medicin e group, with significant difference statistically significant(P<0.01), honeysuckle stem middle dose honeysuckle stem in high dose group comparison between way there was no significant statistical significance(P>0.05), but compared with honeysuckle stem l ow-dose group, with significant difference statistically significant(P<0.01); Honeysuckl e stem compound with western medicine can reduce the model rats of articular cartila ge IL-1β expression, there are significant difference compared with model group, the s tatistical significance(P<0.01), honeysuckle stem compound group compared with the western medicine group, with significant difference statistically significant(P<0.01), ho neysuckle stem middle dose group compared with blank group, there was no statistical ly significant difference(P>0.05), honeysuckle stem middle dose group, honeysuckle st em in high dose group is compared between two groups have no significant statistical significance(P>0.05), compared with honeysuckle stem low-dose group, but there are significant difference(P<0.01).Conclusion:Honeysuckle compound can reduce the swelling degree hyperuricemia model rats serum uric acid and acute gouty arthritis model rats, improve the model of rat gait, reduce joint pain and inflammation, its anti-inflammatory swelling and pain of GA curative effect is probably by inhibiting IL-1β, and high expression of TNF-α and play a role. |
