Objective : The reverse transcription polymerase chain reaction(RT-PCR) and matrix-assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS) techniques were used to explore the FOSL2 gene expression and DNA methylation state of Xinjiang Uygur type 2 diabetes mellitus(T2DM)patients respectively.Methods: 1. The blood samples and general data from 50 cases T2 DM and 50 cases non DM of Uygur in Shache County Xinjiang were collected from the year of November 2013 to January 2014. Then the samples were anticoagulated by the heparin, and the sex,year,weight,height,waist,hip,blood pressure and course of diabetes were measured and record, then calculate the body mess index(BMI) and waist hip radio(WHR).Fasting plasma glucose(FPG) and lipids were simultaneously detected by automatic chemistry analyzer. The level of fasting insulin(FINS)was tested by chemiluminescent immunoassay. The level of glycosylated hemoglobin(Hb A1c)was measured by high pressure liquid chromatography(HPLC).2. The expression of the FOSL2 gene was detected by the RT-PCR. 3. The level of the DNA methylation of FOSL2 gene from the peripheral blood leukocyte was analyzed by MALDI-TOF MS method.4. All the statistical analyses were performed using the SPSS 17.0 software. All P-values presented were two-tailed.P<0.05 was considered to indicate a statistically significant difference.Results: 1.Compared with Uyghurs NDM group,the clinical basic data,such as age,sex radio, DBP, SBP,WHR had no statistically significant differences in Uyghurs T2 DM group(P>0.05).2.Compared with normal control group,the m RNA expression levels of FOSL2 gene in Xinjiang Uygur T2 DM patients group were significantly lower(0.996 ± 0.159,2.456 ± 0.512; P <0.01). 3. Compared with Uyghurs NDM group,the methylation rates of CPG Unit3(CPG3),CPG Unit4(CPG4.5),CPG Unit5(CPG6.7),CPG Unit6(CPG8),CPG Unit8(CPG11),CPG Unit9(CPG12.13.14),CPG Unit10(CPG15.16.17) of FOSL2 gene in Xinjiang Uygur T2 DM patients group were higher. 4. In the multiple linear regression analysis, the expression of FOSL2 gene level as dependent variables, the levels of age,year,weight,height,waist,hip,WHR,BMI,SBP,pulse rates, SBP,DBP, FPG,Hb A1 c,TC,TG, and other indicators as independent variables,the results displayed height, weight, BMI,SBP,FPG and Hb A1 c went into the regression equation,that reminder they may be the main factors of influencing FOSL 2 gene expression level. The regression equation is y=-1.521+0.007(Height)-0.006(Weight)+0.018(BMI)+0.001(pulse rate)+0.001(SBP)-0.003(FPG)+0.004(Hb A1c).Conclusion: 1.FOSL2 gene may be associated with the Xinjiang Uygur T2 DM, obesity and abnormal glucose and lipid metabolism. 2.The hypermethylation of several DNA sites of FOSL2 could lead to the down-regulation of m RNA expression levels, involved in the occurrence and development of Xinjiang Uygur T2DM。...
