Objective:By comparing the acute toxicity test,efficacy test and subacute toxicity test of "toxic" asteraceae,pungent drug Xanthii and "non-toxic" Asteraceae pungent drug Siegesbeckiae Pharmacopeia recorded. To determine the toxicity effect relationship and to provide clinical medication safety precautions,lay the foundation for the definition and classification of Chinese medicine toxicity.Methods:Prepare decoction powder of Xanthii and Siegesbeckiae.(1)Acute toxicity study using fed a one-time in mice to observed toxicity and Calculate the median lethal dose;(2)Efficacy study using ice composite adrenaline common to copy Rat Model of Acute Blood and to investigate the effects of the two drugs on rats with acute blood hemorheology by measuring whole blood viscosity,plasma viscosity,hematocrit,erythrocyte aggregation index,erythrocyte rigidity index and erythrocyte deformation index and to observe the effects on coagulation function in rats of the two drugs by measuring prothrombin time,thrombin time,activated partial thromboplastin time and plasma fibrinogen levels;(3)Subacute toxicity test using fed the two drugs in rats to determine Siegesbeckiae and Xanthii organ damage in rats and to observe the extent of damage to the organs of rats and to observe the toxicity relationship between them by measuring blood Routine,blood biochemical parameters,and pathology;(4) By measuring SOD and MDA on the organ,and explore the mechanism of toxicity of both drugs.Results :(1) Acute toxicity test results:Median lethal dose of Siegesbeckiae LD50=18.0g?Kg-1(Equivalent to the original drug 186.14g?Kg-1);Median lethal dose of Xanthii LD50=18.6g?Kg-1(Equivalent to the original drug 194.15g?Kg-1).There was no significant difference between the two median.(2)Efficacy test results:Compared with the normal group, the model group rats’ Hemorheology were significantly increased;Prothrombin time and activated partial thromboplastin time was significantly shorter,Plasma fibrinogen levels were significantly increased.Compared with the model group,Aspirin group,Siegesbeckiae group and Xanthii group could reduce blood rheology indexes,prolonged the prothrombin time and activated partialthromboplastin time Significantly,Plasma fibrinogen levels decreased significantly. Toxic dose efficacy dose relations ratio of Siegesbeckiae is 85 times,and the Xanthii is 75 times,the toxic effect relationship between Siegesbeckiae and Xanthium is similar;(3)Subacute toxicity test results show,there is damage to rats’ liver and there exists dose-effect relationship,after stopping drugs two weeks compared with the control group,the difference is reduced,indicating that the toxicity of Xanthii is reversible.Siegesbeckiae have damaging effects on the lungs,the dose is high,serious damage, after stopping drugs two weeks the difference significantly reduced compared control group,indicating that the toxicity of Siegesbeckiae is reversible too.(4)For 4 weeks,Xanthium group decreased liver SOD,MDA significantly increased.Siegesbeckiae decreased lung SOD,MDA significantly increased.Conclusion:(1)Xanthium accord to four items “Severe symptoms of poisoning,liver damage,a larger amount of death,poisoning incubation period of 10-30min”of Multi-index gradings Consistent with the Chinese Pharmacopoeia records "Xanthii is toxic drug, "(2) Siegesbeckiae also consistent with four items of multi-index stagings“Severe symptoms of poisoning,liver damage,a larger amount of death,poisoning incubation period of 10-30min”.Siegesbeckiae is "toxic" medicine or not.(3)Comprehensive analysis of the experimental results suggested there exists inadequate in defining and classification of toxic medicine in Pharmacopoeia,to providing precautions to safety clinical medication,and to lay the foundation for the definition and classification of Chinese medicine toxicity.
