Objective:Sarcococca ruscifolia Stapf.belongs to Sarcococca in family of Buxaceae.We found that the total alkali of Sarcococca ruscifolia had a better quality of pharmacological activity.But,the toxicity of Sarcococca ruscifolia need to be evaluated.Consequently,we conducted the acute toxicity test,subacute toxicity test,and the long term toxicity test to explore the pharmaceutical value to insure the medication safety.Methods:We used intragastric administration in animal experiment.The LD50 value was measured by Karher’s Method,and we observed the behavior paremeters by eyesight.The weight,food ration,and water consumption were measured in a set time.We measured the weight of viscera and calculated the organ coefficient.The biochemical criterion of blood was measured by fully automatic biochemical analyser,and the pathological histology slices was dyed by Hematoxylin Eosin(HE)and was observed by fluorescence microscope.In addition,the severity of histopathologic changes was measured by semi-quatitatice method.Finaly,we used SPSS software to statistic the the experimental data.Results:All results were below:1.Acute toxicity testThe LD50 of the total alkali of Sarcococca ruscifolia was 237.69mg·kg-1,and the 95%confidence limit was from 202.05mg·k-1 to 279.62mg·kg-1.We observed that the mice had poor spirit,dim clothing hair,unstable,tail-raising,and convulsion in abdomen,and had high mortality.We also found that the weight of experiment groups was lower less than control groups(p<0.05).The results suggested that the livers were mainly in edema,and some kidney tubles were also in edema,yet all mice had normal cardiac muscle tissue.2.Subacute Toxicity test(1)We found that the mice in high-dose and medial-dose groups were in poor spirit,had dim clothing hair,and diarrhea.Furthermore,we found three mice died in high-dose group,and two mice died in medial-dose group.(2)We found that the weight of the mice in high-dose group were significantly different from that in control group during one to four weeks,while the weight in medial-dose group were significantly different from that of control group at first week.We observed that the concentration of creatinine,glutamic-pyruvic transaminase were significantly higher than that of control group,while the concentration of total bilirubin was significantly less than that of control group.In the medial-dose group,the results showed that the concentration of glutamic-pyruvic transaminase was significantly higher than that of control group.In addition,the concentration of total bilirumin was significant less than that of control group.(3)We observed that the liver impairment was changed with gradient,in high-dose group>in medial-dose group>in low-dose group,of which some liver tissure was in edema.We also found that some kidney tubles had a little edema,while we did not observe pathological changes in cardiac tissure,spleen,lung,and brain.3.Long term toxicity test(1)We found that the mice in high-dose and medial-dose groups had a poor spirit,bad clothing hair,diarrhea et al.In addition,we observed nine mice died in high-dose group,and one mouse died in medial-dose group.(2)The water-intake of mice in high-dose group in mid-experiment was significantly higher than control group,while the mice in medial-dose group was significantly lower than that of control group at the fourth week.At the end of experiment,the water-intake in high-dose group was significantly lower than that of control group,while the water-intake in medial-dose group was significantly lower than control group at the seventh,tenth and eleventh week.The water-intake in mice of all experiment groups were almost the same with that of control group.(3)The food ration of the mice in high-dose group in the mid-experiment was significantly lower than that of control group,and the food ration of the medial-dose group was significantly lower than that of control group at the second and the third week.The food ration of the mice in high-dose group was significantly lower than that of control group.In addition,the food ration of mice in recovery phase of all group had no significant difference.(4)The weight of mice in high-dose group in mid-experiment was significant lower than control group,the mice in medial-dose group was significantly higher than control group at two to four weeks,and the mice in low-dose group was significantly less than control group at second week.The weight of mice in high-dose group at the end of the experiment was significantly lower than control group.In addition,we observed no difference of weight in the recovery phase.(5)We observed that the concentration of creatinine,glutamic-pyruvic transaminase,total bilirubin,and triglyceride in high-dose group were significantly lower than control group.And the concentration of creatinine and total bilirubin were significantly lower than that of control group.The concentration of glutamic-pyruvic transaminase,glutamic oxalacetic transaminase,total bilirubin,triglyceride,and glucose were significantly lower than that of control group,while the concentration of the creatinine was significantly higher than that of control group.The concentration of creatinine,glutamic-pyruvic transaminase,and triglyceride were significantly lower than that of control group.In addition,the concentration of the creatinine was significantly lower than control group.We did not observe any difference of biochemical criterion between all groups and control groups.(6)The haematological index of all groups have no difference compared with control group,while we did not find significant difference between all groups and control groups in the concentration of thrombocyte.(7)The organ coefficients of mice in the high-dose group(cardiac,liver,spleen,lung,kidney)were significantly higher than control group,the organ coefficients(lung,kidney)in the medial-dose were significantly higher than control group.We found that the organ coefficients in the low-dose group were significant higher than control group.We observed organ coefficients(liver,lung,kidney)in the high-dose groups were significantly higher than control group in the end of experiment,and we also found organ coefficients(lung,kidney)were significantly higher than control group.We did not observe any difference of organ coefficients between all groups and control group in recovery phase.(8)The kidney tubles had a little edema in the mid and end of the experiment,while they recovered in the recovery phase.Likewise,the liver had a little edema in the mid and end of the experiment,and they recovered in the recovery phase.In addition,we did not find any pathology in brain at any different phases.Conclusions:Our study found the LD50 of the total alkali of Sarcococca ruscifolia was 237.69mg·kg-1,and the 95%confidence limit was from 202.05mg·k-1 to 279.62mg·kg-1.We also found the toxicity to kidney and liver were enhanced as dosage increased,but the toxicity could recover without any alkali.In addition,we did not find any toxicity to cardiac,spleen,lung,and brain.
