Extraction Total Alkaloids From Radix Polygalae And Prepartion Of Its Micospheres

Objective:The study on the technology conditions of total alkaloids of Radix Polygalae extraction& purification, and the research of total alkaloids of Radix Polygalae cancer cytology 。At the same time, preparation conditions of new dosage forms of total alkaloids of Radix Polygalae microspheres also are studied to increase availability of the total alkaloids of Shanxi Polygalae tenuifolia.Methods:Total alkaloids from Radix Polygalae is extracted of by solvent extraction, separated by macroporous resin, used solution with bromocresol green colorimetry to determine the content of ornidazole. For its activity, this research has carried on the experimental study of cancer cytology. Finally, the text try to use single emulsion-drying in liquid method to preparate total alkaloids of Radix Polygalae microspheres, study the best preparation conditions, and estimate its morphological characteristics, particle size, encapsulation efficiency.Results:Using single factor test and method validation methods to determinate of content of total alkaloids of Radix Polygalae: chloroform 10 mL,buffer solution of pH=2.5 8 mL, bromocresol green solution 4 mL were added separatory funnel. Adding proper amount of analyte solution, funnel shake for 2 minutes, standing for 15 minutes, then taken out the chloroform layer. Solution will add to Erlenmeyer flask with plug with the containing anhydrous sodium sulfate 2 g, determined the absorbance at the wavelength of 419 nm. The standard curve equation is: C=86.263A-2.613. Through the polygala root alkaloid content determination, method about determining the extraction of total alkaloids from Radix Polygalae tenuifolia : take coarse powder for 200 g, place in a 2000 mL round bottom flask, add 1000 mL petroleum ether(60 to 90 DEG, 70 DEG C) heating reflux extraction for 2 hours, the operation need repeated 3 times; after extraction of drug residue by 0.1% HCl 1500 mL immersion overnight, filtered impregnating solution; the filter residue is added with water percolation filtration, leachate and the merger of impregnating solution; the merged liquid with 400 g 732 cation exchange ion exchange resin, resin exchange after washing with distilled water until no impurities, dried; after concentrated ammonia is used to mix the wet, waiting for 20 minutes, extraction with chloroform in cable’s extractor for many times, recycling chloroform solution, the total alkaloids of Radix Polygalae chloroform solution is obtained.Research on cancer cell experiment, total alkaloid harmaline harmane alkali and Radix Polygalae on human cervical carcinoma(Hela) cells, human colon carcinoma(HT-29) cells and human gastric carcinoma(BGC-823) cell inhibition strength: Harman alkali > Polygala total alkaloid harmaline,>Luo Tuopeng base, Harman alkali and Radix Polygalae the total alkaloid on human cervical carcinoma(Hela) cell is the strongest of growth inhibition.Polygala total alkaloids of microsphere preparation study obtained 30%PLGA, 4% Twain-80 aqueous solution according to the oil-water ratio is 1/300 prepared Polygala alkaloid morphology of microsphere is best, the grain size of 30.89 + 3.01 m, loading(1.56 + 0.33)%, the entrapment efficiency was(40.46 + 3.23)%.Conclusion:A preliminary study about the extraction & separation of the total alkaloids of Radix Polygalae, study on cell in vitro activity and microspheres preparation, and optimize the experimental conditions, ultimately determine the preparation process conditions of extracting separation method and microspheres.
