Cognitive Dysfunction(CD)refers to the weakening of individual Cognitive functio ns such as learning and memory,attention and executive ability.The main clinical manifestation of cognitive dysfunction is weakened learning and memory function.Polygala tenuifolia Willd.and Polygala sibirica L.is Polygalae Radix of dried root.The method of use is to remove or not remove the wood core.It has educational function and is used fordiseases such as forgetfulness.Processed Polygalae Radix also has the effect of enhancing intelligence.Ancient and modern medicalguide contains Polygalae Radix as the king’s medicine smart soup,can attending forgetful.Therefore,this article chooseed model of cognitive dysfunction induced by scopola mine in mice.Morris water maze and open fieldtest were used to observe the behavioral indexes of mice.The cholinergic indexes such as AchE and ChAT,infla mmatory indexes such as TNF-α and IL-1β,and antioxidant indexes such as SOD,MDA and GSH-Px were determinedby ELISA test.HE staining was used to observecell apoptosis.Compare and verify the effect of Polygalae Radix and its co mpound Smart Soup on improving learning and memory function.The main research results of this paper are as follows:1.Compare different specifications of Polygalae Radix to improve learning and memory effect.According to the clinical human dose of Polygalae Radix tube(10 g/d).The dose of Polygalae Radix mice with different specifications was1.8 g/kg.Polygalae Radix tube and root was able to significantly reduce the levels of AchE,TNF-α and IL-1β levels and increase ChAT.The levels of MDA and SOD and GSH-Pxwere significantly decreased by rhizome,root and seedling.It is suggested that tube,root and seedling can improve the learning and memory effect.The effect order is tube > root > seedling.The water extract of tube,root and se edling can reverse the hippocampal lesion.Rhizome can’t reverse cognitive impairment.2.Compare the effects of Polygalae Radix,Processed Polygalae Radix and Polygalae Radix-Glycyrrhizaev Radix et Rhizoma(100:6)to improve learning and memory.The experimental dose was set according to the dose of Polygalae Radix mice(1.8 g/kg).Compared with Polygalae Radix,Processed Polygalae Radix and Polygalae Radix-Glycyrrhizaev Radix et Rhizoma(100:6)behavioral indicators were significantly improved.The levels of AchE,TNF-α,IL-1β and MDA were decreased,and the levels of ChAT,SOD and GSH-Px were increased.They could reverse the hippocampal lesions.The results showed that all three improved learning and memory.Polygalae Radix > Processed Polygalae Radix > Polygalae Radix-Glyc yrrhizaev Radix et Rhizoma(100:6).3.Verify the effect of Smart Soup on improving learning and memory function.According to the clinical dose of 15 g/d for human,the medium dose for mouse experiment was calculated,and the low(1.35 g/kg),medium(2.7 g/kg)and high(5.4 g/kg,equivalent to tube 1.8 g/kg)dose groups were set.The results showed that lowdose,medium dose and high dose could improve behavioral indexes and reduce theproportion of abnormal cells.Medium and high dose can significantly regulate the level of cholinergic,inhibit inflammatory response,improve antioxidant capacity.It shows that Smart Soup can improve learning and memory in a dose-dependent manner.
