Aim:Establishment of radioactive lung injury model and discusses Jinyu Granules on the treatment of radioactive lung injury model in mice, and according to the variation in content of related cytokines from plasma explore Jinyu Granules mechanism for the treatment of radioactive lung injury.Methods: â‘ To establish animal model:Through the first part of the experiment to determine the C57BL/6 male mice radioactive lung injury model best radiation dose. (2)Through the second part of experiment to explore the treatment Jinyu Granules: Select 120 male mice, in addition to the 30 of Blank control group, all the rest of the mice to accept 17.5 Gy ray mode. Give the model control group (30, saline lavage), positive control group (30, prednisone acetate suspension liquid lavage), Jinyu Granules group (30, Jinyu Granules suspension liquid lavage) corresponding lavage treatment after building, respectively, at 2,4,6 months time points were randomly take 10 mice in each group to death and take the lung tissue. From the body weight in mice, macroscopic observation pulmonary gross morphology, lung coefficient, lung tissue HE and Masson staining results evaluation therapeutic effect in many aspects. â‘¢Through the third part of experiment explore its mechanism of action:After collecting the experimental mice blood, take the upper plasma after centrifugation, liquid chip method measuring the proinflammatory cytokines in plasma of interleukin 6 (1L 6) and interleukin 1 alpha (IL-1 alpha), interleukin 2 (IL-2), tumor necrosis factor alpha (TNF alpha), as well as promoting fibrosis cell factor, transforming growth factor beta 1 (TGF-beta 1) expression. Make a statistical analysis on relevant indicators, explore Jinyu Granules mechanism for the treatment of radioactive lung injury.Results: â‘ Successfully established radioactive lung injury animal model and the most dosage were determined. â‘¡Jinyu Granules therapy effect:In 2,4, and 6 months after building, building groups of mice compared with blank control group mice body weight decreased significantly (P< 0.01). Compared with model control group in 2,4, and 6 months, positive control group and Jinyu Granules group mice body weight were significantly higher (P< 0.01). Jinyu Granules group and positive control group after 2 months no difference (P> 0.05),4,6 month Jinyu Granules group than the positive control group higher significantly (P< 0.01).Building groups of mice lung coefficient of 2,4, and 6 months than the blank control group increased significantly (p< 0.01), model control group increased the most obvious. Jinyu Granules group than the positive control group decreased significantly (p< 0.01). Building groups of mice lung tissue inflammation and fibrosis score in 2,4, and 6 months compared with blank control group were significantly higher (p< 0.01), model control group increased the most obvious. Jinyu Granules group and positive control group no significant difference in 2 months, when in 4 and 6 months, Jinyu Granules group significantly lower than the positive control group(p< 0.01). â‘¢Jinyu Granules mechanism aspects:Building groups of 2,4, and 6 months in mice plasma IL-6, IL-1 alpha,1L-2 and TNF-alpha and TGF-beta 1 expression significantly increased compared with blank control group (p< 0.01),the model control group increased most obviously. Jinyu Granules in mice plasma IL-1 alpha, IL-2 and TNF-alpha and TGF-beta 1 expression in each time point is lower than the positive control group (p< 0.05 or p< 0.05), Jinyu Granules group of interleukin 6 (IL-6) in the first 2 months and the positive control group no difference, in 4,6 month than the positive control group decreased significantly (p< 0.01).Conclusion: â‘ Under certain conditions in mice to 17.5 Gy dose radiotherapy can create radioactive lung injury animal models.â‘¡Jinyu Granules in terms of improvement of radioactive lung injury in mice body weight in mice, lung tissue morphology under the naked eye, lung coefficient and lung tissue pathological changes has more effect than prednisone acetate. â‘¢Jinyu Granules can reduce proinflammatory factor in mice plasma levels of cytokines and alveolar interval hyperemia, hemorrhage and neutrophil infiltration, and thus improve the inflammatory response in lung tissue, and reduce the fibroblast proliferation and differentiation at the same time, reduce the synthesis of collagen, improve lung fibrosis.â‘£Jinyu Granules on the improvement of radioactive lung injury in mice and reduce proinflammatory and promote fibrosis cytokine levels in mice plasma reflects its advantages, particularly long-term effect is better than that of prednisone acetate therapy, can be used in the clinical observation for the treatment of radioactive lung injury.
