Objective:By observing the Tongluotangtai Compound Recipe in type 2 diabetic GK spontaneously sciatic nerve in rats IGF-1 expression, discussion on this side of nerve growth factor and apoptosis aspects of diabetic peripheral neuropathy possible mechanism to provide a theoretical basis for clinical treatment.Methods:7-8 weeks of age 50 male GK rats, high fat feed made die after 12 weeks, according to blood glucose were randomly divided into model group, Tongluotangtai Compound Recipe high, medium and low dose group, western medicine group(metformin+Methycobal), each group of 10. Match up weeks of age, body weight of male Wistar rats 10 for the normal group, a total of six groups. Normal group and model group were given saline 5.0ml/kg.d gavage, Tongluotangtai Compound Recipe high, medium and low dose group were given Tongluotangtai extract 2.88g/kg.d,1.44g/kg.d,0.72g/kg.d gavage, metformin medicine group received 100mg/kg.d and Methycobal 0.2mg/kg.d suspension made gavage, once a day. Observe the general condition of rats, body weight and blood sugar during the administration.28 days after the death, to detect the expression of IGF-1mRNA sciatic nerve and Caspase-3 positive cells and nerve cells apoptosis, rat sciatic nerve conduction velocity and observe its pathological changes.Results:Compared with normal group, model group GK rats blood glucose significantly increased (P< 0.01), sciatic nerve conduction velocity slowed significantly (P<0.01), IGF-1mRNA expression is reduced, Caspase-3 expression increases, the number of nerve cell apoptosis increased (P<0.01), GF-1mRNA downregulated, Caspase-3 expression was increased, increased neuronal apoptosis (P <0.01、P<0.05),sciatic nerve fascicles membrane breakage is incomplete, the nucleus pycnosis nerve fibers, nerve fiber bundle fracture,part of the liquefactive necrosis of pathological change; After the tongluotangtai Compound recipet reatment of diabetic peripheral neuropathy can promote weight gain GK rats, lower blood sugar levels, improve sciatic nerve conduction velocity and pathological damage, increased levels of IGF-1mRNA inhibit the expression of Caspase-3, reducing neuronal apoptosis, which tongluotangtai high dose group and metformin treatment Methycobal medicine group difference (P<0.05)Conclusion:Tongluotangtai Compound Recipet s can improve glucose metabolism of DPN rats, improve sciatic nerve conduction velocity, reduce nerve tissue pathology damage, improve nerve function, play a role in prevention and treatment of diabetic peripheral neuropathy. The mechanism may be up-regulated the expression of IGF-1mRNA of nerve tissue, and by inhibiting the activation of Caspase-3 decreased neuronal apoptosis, to promote nerve regeneration and the role of anti-apoptotic.
