Objective:The experiment tries to explore a possible mechanism of Tongluotangtai Poly-pharmaceutical treating Diabetic Peripheral Neuropathy by observing the inhibition of neuronal apoptosis, and fanally open up new ideas and methods for the prevention and treatment of diabetic peripheral neuropathy.Methods:56 rats which were male,7-8 weeks old and whose fasting blood glucose were more than or equal to 11.1 mmol/L were selected, and given high rat diet for 12 weeks, so that they became the model rat of spontaneous DPN. The 50 rats which were DPN were randomly divided into model group, Tongluotangtai High-dose group, Tongluotangtai dose group, Tongluotangtai Low-dose group, metformin+methycobal group. Wistar rats with age and sex matched normal group 10 were selected, a total of 6 group. Three Tongluotangtai dose groups were fed different dose Tongluotangtai suspension, the western medicine group was treated with metformin 100mg/kg.d and Mecobalamin 0.2mg/kg.d suspension gavage,l times per day, all groups were sacrificed 28 days after intragastric administration. To observe the general condition of rats, change detection of rat fasting blood glucose, sciatic nerve conduction velocity, Pathology of sciatic nerve, Caspase-3 expression level and apoptosis lever in sciatic nerve.Results:The general situation of Tongluotangtai Poly-pharmaceutical groups significantly improved, with better effect in high group, the blood glucose of rats in every treatment groups showed slowly down, and the effect of the western medicine group is most obvious, but the Chinese medicine groups showed no significant difference (P> 0.05), in the improvement of nerve conduction velocity in DPN rats by gavage for 28 days in the treatment groups, MNCV, SNCV of DPN the sciatic nerve rats were increased, and the high-dose group improved significantly (P< 0.01), in low-dose group, middle-dose group and Western medicine group all could improve MNCV and SNCV, but there was no statistical significance (P>0.05); Tongluotangtai Poly-pharmaceutica can also mitigate the sciatic nerve tissue damage, such as nerve bundle membrane damage, nerve fiber nuclei pyknosis, nerve cell fibers beam fracture, and high-dose group’s effect was more obvious. Tongluotangtai Polypharmaceutical could decreased the sciatic nerve caspase-3 expression level, compared with the model group, treatment groups has significant difference (P< 0.01), and compared between the treatment groups, and sciatic nerve caspase-3 expression level in the high-dose group are more significant than low-dose group and western medicine group (P<0.01,P< 0.05), finally Tongluotangtai Poly-pharmaceutica could inhibition of sciatic nerve apoptosis, the apoptosis of high-dose group rate was significantly lower than that in middle-dose group, the low-dose group and Western medicine group (P< 0.05 or P< 0.05).Conclusions:The possible mechanism for the treatment of diabetic peripheral neuropathy of Tongluotangtai Poly-pharmaceutica could be by decreasing the Caspase-3 expression levels and inhibiting apoptosis of nerve cells.
