| PurposreComparative study on Wntã€Notch signaling pathway molecules in primary breast cancer stem cells and matched lymph nodes metastasis stem cells.Expression and significance of Wnt proteins β-catenin and Cyclin D1 in primary breast cancer and matched lymph node metastases.Methods30 cases of breast invasive ductal carcinoma and matched metastasis lymph nodes were made into single cell suspensions, then breast cancer stem cells were separated from them by immunomagnetic sorting, β-catenin, Cyclin D1 and Notchl gene expressions of Wnt and Notch signaling pathway were detected by real time PCR. The protein of β-catenin and Cyclin D1 was detected by immunohistochemistry in a series of 55 triple-negative breast cancer (TNBC) and matched 21 lymph node metastases, and 55 non-TNBC and matched 25 lymph node metastases.ResultsThe expression of P-catenin in primary breast cancer stem cells and matched lymph nodes metastasis stem cells had statistically no differences (P>0.05); While the expression of Cyclin D1 and Notchl in matched lymph nodes metastasis stem cells were significantly higher than the expression in primary breast cancer stem cells (P<0.01, respectively).The protein of β-catenin and Cyclin D1 was detected by immunohistochemistry in a series of 55 triple-negative breast cancer(TNBC) and matched 21 lymph node metastases, and 55 NON-TNBC and matched 25 lymph node metastases. Results Abnormal β-catenin expression rates in TNBC and non-TNBC primary breast cancer were 52.7%(29/55) and 63.6%(35/55). the positive expression rates of Cyclin D1 were 54.5%(30/55) and 58.2%(32/55),β-catenin and Cyclin D1 in TNBC and non-TNBC primary breast cancer had statistically no differences (P>0.05); abnormal β-catenin expression rates in TNBC and non-TNBC lymph node metastasis were 71.4%(15/21) and 80.0%(20/25), the positive expression rates of Cyclin D1 were 76.2%(16/21) and 88.0%(22/25), β-catenin and Cyclin D1 in TNBC and non-TNBC lymph node metastasis had statistically no differences (P>0.05). Abnormal expression rates of β-catenin in 46 primary breast cancer and matched lymph node metastasis were 54.3%(25/46) and 76.1%(35/46), Cyclin Dl positive expression rates were 63.0%(28/46) and 82.6%(38/46), and β-catenin expression were positively correlated with Cyclin D1 expression (P<0.05).ConclusionCompared with the primary cancer stem cells, CyclinDl and Notch 1 activation in metastasis cancer stem cells are in higher level, which leades to a higher capability of invasion and metastasis, that may be a new therapeutic target.Regardless of the primary breast cancer or matched lymph node metastasis, the expresssin of β-catenin and Cyclin D1 in TNBC and non-TNBC had no correlations. Abnormal expression of β-catenin leads to Cyclin D1 positive expression, and Cyclin D1 positive expression promote primary breast cancer invasion and lymph node metastasis. |
