Objective(1)To investigate the relationship between estrogen receptor (ER) and the expression and clinical significance of Hedgehog signaling pathway molecules Ptch and Glil in the breast invasive ductal carcinoma. (2)To investigate the relationship between human epidermal growth factor receptor-2 (HER2) and the expression and clinical significance of Notch signaling pathway molecules Notch1 and Jag1 in the breast invasive ductal carcinoma.MethodsThe surgical specimens of 120 breast invasive ductal carcinoma patients’ tissues were obtained between the period 2011-10 and 2013-10 in Binzhou Medical College Hospital.The expression and clinical significance of Gli1 and Ptch were examined using Real time-PCR and Western blot in 60 cases of ER-positive and 60 cases of ER-negative specimens taken from patients with complete clinical data.The surgical specimens of 120 breast invasive ductal carcinoma patients’ tissues were obtained between the period 2011-01 and 2013-09 in Binzhou Medical College Hospital.The expression and clinical significance of Notch1 and Jag1 were examined using Real-time PCR and Western blot in 60 cases of HER2-positive and 60 cases of HER2-negative specimens taken from patients with complete clinical data.Results(1)The expression of Glil mRNA was 1.05±0.27 in ER-positive group, and the expression of Gli1 mRNA was 2.70±0.26 in ER-negative group, the differences were statistically significant (t=20.751,P<0.01). The expression of Ptch mRNA was 1.04±0.29 in ER-positive group, and the expression of Ptch mRNA was 2.34±0.35 in ER-negative group, the differences were statistically significant (t=18.413,P<0.01). The expression of Gli1 protein was 0.43±0.007 in ER-positive group, and the expression of Gli1 protein was 0.71±0.011 in ER-negative group, the differences were statistically significant (t=11.086,P<0.01). The expression of Ptch protein was 0.36±0.008 in ER-positive group, and the expression of Ptch protein was 0.58±0.012 in ER-negative group, the differences were statistically significant (t=16.437,P<0.01). The expression of Glil and Ptch were associated with disease stage and metastasis (all P<0.05),but no correlated with tumor size and ages(all P>0.05)There was positively correlation between Glil and clinical staging (χ2=12.17, P<0.01).There was positively correlation between Ptch and clinical staging (χ2=7.88, P<0.01).There was positively correlation between Glil and lymphatic metastases (χ2=6.41, P< 0.05).There was positively correlation between Ptch and lymphatic metastases (χ2 =5.92,P<0.05).(2)The expression of Notchl mRNA was 1.12±0.21 in HER2-positive group, and the expression of Notchl mRNA was 2.64±0.34 in HER2-negative group, the differences were statistically significant (t=18.437,P<0.01). The expression of Jagl mRNA was 1.04±0.23 in HER2-positive group, and the expression of Jagl mRNA was 2.43±0.27 in HER2-negative group, the differences were statistically significant (t=16.521,P<0.01). The expression of Notchl protein was 0.40±0.008 in HER2-positive group, and the expression of Notchl protein was0.65±0.007 in HER2-negative group, the differences were statistically significant (t=10.126,P<0.01). The expression of Jagl protein was 0.25±0.007 in HER2-positive group, and the expression of Jagl protein was 0.67±0.005 in HER2-negative group, the differences were statistically significant (t=14.353,P<0.01). The expression of Notchl and Jagl were associated with disease stage and metastasis (all P<0.05),but no correlated with tumor size and ages(all P>0.05)There was positively correlation between Notch and clinical staging (χ2=8.83, P<0.01).There was positively correlation between Jag1 and clinical staging (χ2=5.86,P<0.01).There was positively correlation between Notch1 and lymphatic metastases(χ2=12.6, P<0.01).There was positively correlation between Ptch and lymphatic metastases (χ2=12.6,P<0.01).Conclusion(1)The expression of Gli1 and Ptch significantly higher in the ER negative breast cancer, The expression of Gli1 and Ptch had positive correlation with ER negative breast cancer, The expression of GLi1 and Ptch may be associated with clinical staging, axillary lymph node metastasis, thus blocking the Hedgehog signaling pathway may become a novel targets for the treatment of ER negative breast cancer.(2)In the HER2-negative breast cancer, Notch signaling pathway is significantly activated, The expression of Notchl and Jag1 may be associated with clinical staging, axillary lymph node metastasis,blocking the Notch signaling pathway may become a new therapeutic target for HER2-negative breast cancer.
