Objective:To evaluate the relationship between vitamin D receptor and genetic susceptibility to overweight/obesity in school-age children of Han and Kazakh in Xinjiang Uygur Autonomous Region. Methods: The VDR gene genotypes were detected by using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method in 250 overweight/obesity school-age children(127Han,123Kazakh) and 255 healthy controls(131Han,124Kazakh),and analyze the distribution of alleles and genotypes of the BsmI、TaqI and ApaI sites of the VDR gene. Results:(1) Genotypes frequency distribution of the BsmI 、 TaqI and Apa I sites of the VDR gene are all accord with HWE(Hardy-Weinberg equilibrium)(P>0.05),and explains the research sample is representative.(2)There was statistically significant difference in the frequencies of three SNPs genotypes and alleles between the healthy controls of Han and Kazakh(P<0.05).(3)There was statistically significant difference in the frequencies of ApaI between the overweight/obesity cases and healthy controls of Han(P<0.05). AA/Aa+aathe difference in frequencies of ApaI between the overweight/obesity cases and controls of Kazakh was statistically significant(P<0.05);There was no statistically significant difference in the frequencies of BsmI between the overweight/obesity cases and healthy controls of school-age children of Han(P>0.05),but there was statistically significant difference in female school-age children of Kazakh(P=0.048);There was statistically significant difference in the frequencies of TT/Tt+tt genotype and the alleles between the overweight/obesity cases and healthy controls of Kazakh(P<0.05).But the difference in frequencies of TaqI genotypes and alleles between the overweight/obesity cases and controls of Han was not statistically significant(P>0.05).There was no statistical differences in combined effects of any two gene loci between the Han and Kazakh school-age children in the two groups(P>0.05).There was statistical differences in combined effect of the three gene loci between the cases and controls in Han school-age children(P=0.01).There was no statistical difference between the two groups in Kazakh school-age children(P=0.06).Conclusion: VDR gene SNPs polymorphism was exsit ethnic differences.The polymorphisms of Apa I are associated with the susceptibility to overweight/obesity in school-age children of Han,while there is not association with the susceptibility to overweight/obesity school-age children in Kazakh;The polymorphisms of BsmI are not associated with the susceptibility to overweight/obesity in school-age children of Han,but there is association with the susceptibility to overweight/obesity school-age children in Kazakh;The polymorphisms of TaqI are not associated with the susceptibility to overweight/obesity in school-age children of Han,while there is association with the susceptibility to overweight/obesity school-age children in Kazakh,especially male school-age children.There may be no obvious correlation with the combination of any two gene locus in Han and Kazakh overweight/obese school-age children.The combined effects of three sites may be associated with the han overweight/obese school-age children,but There was no obvious correlation with Kazakh overweight/obese school-age children.
