Objective:Optic neuritis, a common disease in ophthalmology, is attributed to the category of “sudden blindness of ocular connectors†in traditional Chinese medicine. It is convinced that the stagnation of liver Qi is the most common cause about the disease. This study is aimed to provide guidance to clinical practice by observing the dynamic variation of-D-N-methyl-aspartate receptor1( referred to NMDAR1 or NR1) in prepared rats model with optic neuritis caused by liver qi stagnation.Methods:Building the rats model with optic neuritis caused by stagnation of liver qi by copying the method of introducing chronic unpredictable stimulation to them while combining the experimental autoimmune encephalomyelitis(EAE experimental autoimmune encephalomyelitis) model introduced by CNS(central Nervous system, CNS) to investigate the impact of the goal to the study by applying xiaoyaosan, the representative prescription of conditioning liver, to conduct pharmacy intervene. Then, by observing the morphalogical changes of optic nerve and retina from the level of organ and the temporal and spatial change of visual cortex in the visual pathway and the lateral geniculate NR1 from the level of protein to provide evidence to prove the close relationship between liver and the ocular connectors.Results:Compared with the normal group, the rats with neuritis caused by liver qi stagnation, which have received chronic unpredictable stimulation combining with the antigen stimulation introduced EAE model modeling for 28 days, have shown the symptom of weight deduction, lasititude, curls up with limp limbs, and eat less; besides, some of them become strengthless, or even suffers paralysis. Meanwhile, the sugar intake of them has been deduced; in addition, the dense of ACTH in plasma has surged, the liver has been changed organically and the retinal changed pathologically. While, comparing the rats with neuritis caused by liver qi stagnation with those in control group after being immunohistochemical stained, the NR1 in visual cortex and the lateral geniculate in protein expression have increased simultaneously. On the contrary, the NR1 in visual cortex and the lateral geniculate in protein expression have decreased after being stained as the same way above.Conclusion:In this study, the prepared rats model with neuritis caused by liver qi stagnation which is made by unpredictable chronic stimulation combining with antigen induced EAE model is successful. The increased expression of NR1 protein proves that the stagnation of liver qi is the factor that affects the function of the eyes to the rats with neuritis caused by liver qi stagnation. While the decreased expression of NR1 protein in the group of rats with neuritis caused by liver qi stagnation intervened by xiaoyaosan shows that xiaoyaosan, the representative prescription of conditioning liver, has great improvement to the eye-function of rats with neuritis caused by liver qi stagnation. |
