| Objective : Through the experiments of Xiaoyao Powder group liver qi stagnation syndrome model rats of optic neuritis,qi stagnation in optic neuritis N-methyl-Daspartate receptor 1(NMDR1,NR1),beta amyloid precursor protein(beta-amyloid precursor protein,referred to as the beta-APP)dynamic changes of ultrastructure and visual pathway change,and change of Xiaoyaosan intervention in these indicators,to discuss the possible mechanism of Xiaoyao Powder in treating liver qi stagnation syndrome of optic neuritis,and provide laboratory evidence for clinical diagnosis and treatment of optic neuritis and provide scientific basis for clinical treatment.Methods:60 Wistar healthy rats by RAND function were randomly divided into three groups,including normal group,qi stagnation of optic neuritis group and Xiaoyao San plus qi stagnation group of optic neuritis,establishment of liver qi stagnation syndrome animal model with chronic unpredictable stress stimulation method,induced guinea pig spinal cord homogenate antigen test the allergic encephalomyelitis model on the foundation of the card(Experimental allergic encephalomyelitis,referred to as EAE)method to establish the animal model of optic neuritis animal model,combined with the establishment of animal model of disease and syndrome.Then select Tiaogan representative prescription Xiaoyaosan drug intervention.The normal group and qi stagnation group(combination group referred to as optic neuritis syndrome)with normal saline,Xiaoyaosan plus liver qi stagnation group(referred to as optic neuritis Xiaoyaosan group)with Xiaoyao Powder orally,treatment for 28 days.To observe the behavior of rats school,and recorded the rats optic nerve function score,open field test,the morphological change from tissue and organ morphology under light microscope,liver,adrenocorticotropic hormone ELISA method for detection of plasma of rats(Adrenocorticotropic hormone,referred to as ACTH)content change And using the index evaluation model after the success of modeling,immunohistochemical method to detect NR1 in the visual cor tex,the dynamic changes of beta-APP NR1 in the lateral geniculate nucleus and optic nerve were observed by transmission electron microscope,visual cortex,lateral geniculate nucleus,the ultrastructure of the optic nerve.Results:1.Preparation of the model group.Combining disease with syndrome,Xiaoyaosan group rats by chronic unpredictable stress stimulus method combined with the EAE model of antigen induced twenty-eighth days after modeling,compared with the normal group,rats,listlessness,combined with disease group and disease diet reduced,limb movement disorder,even paralysis,optic nerve the function score decreased;disease group and combination of disease and syndrome score of experimental group rats in open field decreased;the disease group and the combination of disease and syndrome with plasma concentration of ACTH rats increased;group under the light microscope,liver pathological changes of the combination of disease and syndrome;animal model.2.Immunohistochemistry group.Combining disease with syndrome: compared with normal group,NR1 in the visual cortex,lateral geniculate nucleus NR1,optic nerve beta-APP protein expression were increased,with statistical significance(P < 0.05).Visual cortex NR1 protein for twenty-eighth days compared to fourteenth days and increased expression of NR1 protein expression in the lateral geniculate body twenty-eighth days is fourteenth days decreased expression of optic nerve beta-APP twenty-eighth days compared to fourteenth days and increased expression were statistically significant(P < 0.05).Xiaoyaosan group: compared with the combination of disease and syndrome group,NR1 in the visual cortex,lateral geniculate nucleus NR1,optic nerve beta-APP protein expression decreased,with statistical significance(P < 0.05);compared with the normal group,NR1 in the visual cortex,lateral geniculate nucleus NR1,optic nerve beta-APP protein expression were increased,with statistical significance(P < 0.05).Combined with the time change with group.3.Ultrastructure under transmission electron microscope.The combination of disease and syndrome rats visual cortex and LGN neurons,changes in glial cells and syna pses were pathology,optic nerve myelin showed clear layers of onion like changes,visual cortex and optic nerve pathological changes with time increased,the lateral geniculate body ultrastructure is fourteenth days twenty-eighth days of serious;Xiao yaosan intervention,Xiaoyaosan group rat lateral geniculate body and visual cortex ultrastructure results were improved,optic nerve myelin dense order,fourteenth days and 28 days more than three visual parts of the recovery results with combination of syndrome group.Conclusion: 1.This experiment used unpredictable chronic stimulation stress method combined wi th EAE model induced by immune antigen.The model of optic neuritis in rats was successful.2.From the site of action,Xiaoyao Powder can be regarded in visual cortex of rats with the way to reduce disease and syndrome,lateral geniculate body and optic nerve NR1 beta-APP expression showed that TGFY Xiaoyao Powder on liver qi stagnation syndrome of optic neuritis rat eye has the function of improving,it is speculated that Xiaoyao Powder by rat visual cortex.Large part NR1,lateral geniculate body and optic nerve beta-APP balance to its treatment of liver qi stagnation syndrome of optic neuritis.From the time,the combination of disease and syndrome group of NR1 in the visual cortex and optic nerve beta-APP increase with the time increase the expression of NR1 in the lateral geniculate nucleus with the increase of time expression decreased,indicating the road The pathological changes of optic cortex,lateral geniculate body and optic nerve in these three sites vary in time,so it is presumed that the order of optic neuritis in the stagnation of liver Qi syndrome may be optic cortex,optic nerve and lateral geniculate body.3.The visual cortex of rats with improved disease Xiaoyaosan,lateral geniculate nucleus,optic nerve ultrastructure,suggesting that Xiaoyaosan can partially restore the ultrastructure of optic pathway through the effect of its treatment of liver qi stagnation syndrome of optic neuritis in rats,the experiment shows that the path of Xiaoyaosan on ultrastructure of rats the first part is the visual cortex may recover. |
