Objective:The research is aimed to observe the impact of yin and yang heel vessels electroacupuncture on the nerve function, motor function, muscle tension and the number of vesicular GABA transporter (VGAT), GABA transaminase (GABA-T) and succinic semialdehyde dehydrogenase (SSADH) expression of cerebral ischemia reperfusion model rats, in order to explore the possible therapeutic mechanism of electroacupuncture on ischemic stroke spastic hemiplegia.Methods:The 30 Sprague-Dawley rats were divided into 5 groups according to the random number table:the empty group, the sham operation group, the model group, the elcetroacupuncture group and the rehabilitation groups. Rats were established to the middle cerebral artery occlusion (MCAO) induced by Zea-Longa method. After three days, the rats were respectively treated by electroacupuncture and rehabilitation training, one time a day and last for 6 days. To evaluate neural function with Bederson Neurobehavioral scores and evaluate movement function with balance beam walk scores and nets screen test at 1,3,9 days after surgery, and obseve the change of muscle tension before and after treatment. After treatment, the rats were killed, removing tissue in clean ice table. The brainstem, intumescentia cerviealis and intumescentia lumbalis were removed and observed with the light microscope. VGAT, SSADH and GABA-T of tissues were detected using Reverse transcription-polymerase chain or ELSIA.Results: 1. Bederson neurobehavioral scores and Motor function scores:before treatment, compared with the empty group and the sham operation group there were no statistical significances between them (P>0.05); compared with the empty group and the model group, the differences were statistically significant (P<0.01), but there was no statistical significance between them (P>0.05). After 9 days of surgery, there were statistical significane (P<0.05) between the model group and and the two treatment groups.2. Using electrophysiological assessment and Ashworth assessment evaluate the muscle tension:before the treatment, compared with the empty group and the sham operation group there were no statistical significance between them (P>0.05); compared with the empty group and the model group, the differences were statistically significant (P<0.01); but there was no statistical significance between them (P>0.05). After 9 days of surgery, there were statistical significane (P<0.05) between the model group and the two treatment groups.3. HE sections:â‘ in the model group, the brainstem showed extensive degeneration, most condensation nuclei, nucleoli were disappeared, cytoplasmic edema, hemorrhage, and inflammatory cell infiltration were found. Blood spots were found in the gary matter neurons of intumescentia cerviealis and intumescentia lumbalis and there were some inflammatory cell infiltration in the glial cells. â‘¡ in the elcetroacupuncture group, brainstem structure intact cells arranged in order, no edema, degeneration and necrosis. And gary matter neurons of intumescentia cerviealis and intumescentia lumbalis were normal, white matter nerve fibers were no degeneration, necrosis and inflammatory cell infiltration.4. The VGAT expression of SSADH in each group using the RT-PCR:compared with the empty group and the sham operation group there were no statistical significance between them (P>0.05); compared with the empty group and the model group, the differences were statistically significant (P<0.01); compared with the model group, the expression of VGAT in the brainstem, intumescentia cerviealis and intumescentia lumbalis was significantly lower, the difference was statistically significant (P<0.05).5. The mean optical density of GABA-T in each group using the ELISA: compared with the empty group and the sham operation group there were no statistical significance between them(P>0.05); compared with the empty group and the model group, the differences were statistically significant(P<0.01); compared with the model group, the expression of VGAT in the brainstem, intumescentia cerviealis and intumescentia lumbalis was significantly lower, the difference was statistically significant (P<0.05).Conclusion:1. Yin and Yang heel vessels electroacupuncture treatment could relieve cerebral infarction body spasms, obviously improved cerebral infarction rats neural function and motor function.2. Yin and Yang heel vessels electroacupuncture treatment may reduce the activity of VGAT in the central nervous system that can make clear reduction of GABA. So that the GABA concentration might be improved to exert the inhibitory synaptic transmission in the synaptic cleft. This could be one of the mechanisms of anti spastic limbs hemiplegic after cerebral infarction.3. Yin and Yang heel vessels electroacupuncture treatment may reduce the activity of GABA-T and SSADH in the central nervous system that can slow down the degradation rate of GABA. Thereby it can improve the concentration of GABA in the synaptic cleft which can play a presynaptic and postsynaptic inhibition effect. It could be one of the mechanisms of anti spastic limbs hemiplegic after cerebral infarction.
