Objective: To observe the effects of Tiao Zang Shu Mi Tang(TZSMT) on the intestinal function in STC model rats, and to investigate the changes of ICCã€SP and VIP in STC model rats, and to investigate the therapeutic effect and possible mechanism of TZSMT for STC.Methods: The SD rats were randomly divided into normal control group and model group. The model group was given compound diphenoxylate suspension to establish STC model. All the model rats were randomly divided into the TZSMT low doseã€middle dose and high dose treatment group, cisapride treatment group and model control group after the STC model rats were made successfully. The normal control group was as same as befor. After completion of therapy, blood samples were collected from heart and then measuring the serum SPã€VIP by ELISA detection. Measuring the intestinal carbon ink propulsion rate and getting the proximal colon tissue specimen after the rats were killed by cervical dislocation,which was to observe the expression of ICC in colon by immunohistochemistry.Results:(1) Compared with the normal control group, vacuolar degeneration was found in ganglion cells in intestinal wall in model group,in which the cells became smaller, shrinkager, uneven dyeing, and had reduced number of neurons and mild swelling of fiber. Therapeutic effect: there was no improvement in TZSMT high dose group. TZSMT low dose group and cisapride group improved obviously, TZSMT middle dose group was at the middle.(2)There was no statistically significant difference between TZSMT low dose group and cisapride group on increase colon advance rate of carbon powderã€value of IOD of ICC in colon and serum SP concentration:(P>0.05); There was no statistically significant difference between TZSMT high dose group and TZSMT middle dose group(P>0.05); TZSMT low dose group and cisapride group were better than TZSMT high dose group and TZSMT middle dose group and the difference was statistically significant(P<0.05).(3)comparising the concentration of VIP in serum: there was no significant difference between the groups(P > 0.05).Conclusion:(1)The colon ICC and serum SP were involved in the pathogenesis of STC as the number of colonic ICC and serum SP were all decreased in STC.(2) Low doses of TZSMT have therapeutic effect to STC rats as promoting intestinel emptying. Probably increasing the number of ICC in the colon wall and the concentration of SP in sarum would be the mechanism of its function,which is better than cisapride.(3)TZSMT would not significantly affect the concentration of VIP in sarum in STC rats.(4)The treatment effect of TZSMT was not proportional to the increase of its does in STC. |
