Comparative Study Of Reproductive Hormone Levels, Histopathology And Expression Of SRB1 And CYP2E1 Of POF Disease Rat Model And Different Uyghur Abnormal Syndrome Rat Models

Objective:To study the connection between POF disease and abnormal Uyghur helits,establish western POF syndrom rat model and different abnormal Uyghur helits rat models,compare the diffefences of hormones,histopathology, SRB1 and CYP2E1,provide the basis for the research of POF in the occurrence,development and Uyghur medicine treatment of POF. Method:100 mature female Wistar rats were selected and were devided into two groups,first group:20 of them were randomly devided into POF group(group A),20 of them were devided to the abnormal sapra group(group B),20 of them were devided to nomal group(group N1),second group,20 of them were devided to abnormal sawda group(group C),20 of them were devided to normal group(group N2).Cisplatin induced POF disease rat model(group A) were established by intraperitoneal injection of CIS.Abnormal sapra syndrome rat model(group B) were established by hot dry environment,dry and heat feed, stimulated by electric shock,noise and tail clamp, according to theory of Uyghur medicine.Abnormal savda syndrome rat model(group C) established by dry and cold environment,dry and cold feed,stimulated by electric shock, braking,and swimming, according to theory of Uyghur medicine.(1)The biological characterization in each group of rats were observed.(2)Reproductive hormone levels of each group were measured by radiation immunization analysis.(3)The morphological changes of hypothalamo-pituitary-ovarian axis were observed by optic microscope. (4)SRB1 and CYP2E1 were measured by immunohistochemical technique. Result:(1)The observation results of biological characterization:Body weight of group A were decreased,feed intake, water intake,weight of urine and stool were decreased,white tongue,hair loose,lethargy,malaise,sluggish,unresponsive.Growth of body weight in group B were retardation,water intake were decreased,water intake were decreased,stool dry, yellow tongue,coarse hair,strong stimulus response,irascibility.Growth of body weight in group C were retardation, feed intake,water intake,weight of urine and stool were increased,stool dry,dark purple tongue,cool body temperature,lethargy,irritability, aggressive,eccentric.(2)Reproductive hormone levels:The levels of E2 in group A and group B were significantly lower than group N1(P<0.05),and the levels of LH and FSH were significantly higher than group N1,(P<0.05),the level of E2 in group C was significantly lower than group N2(P<0.05),the level of FSH was lower than group N2,but no significant differences(P>0.05),the level of LH was higher than group N2,but no significant differences(P>0.05). There were no significant differences in E2, FSH and LH levels between group A and the group B(P> 0.05),but there were significant differences in E2 FSH and LH,levels between group A and the group C(P< 0.05).(3) Morphological changes:Hypothalamus:In the group A,the number of neurons were decreased,and gliocyte hyperplasia.In the group B,the number of neurons were decreased,and arranged irregularly.In group C,gliocyte hyperplasia and swelling,the number of neurons were decreased,the volume was atrophied,the structure was not clear;Pituitary:In the group A,the number of eosinophilic cell were increased,the number of blood sinus were decreased,the volume was atrophied.In the group B,the number of eosinophilic cell were increased,the number of the blood sinus were decreased,and the volume was atrophied.In the Group C,the number of basophilic cell were increased,the number of eosinophilic cell were decreased,the number of the blood sinus were decreased,and the volume was atrophied. Pituitary cell proliferation and were arranged irregularly.Ovary:In the group A,the number of luteal cells were decreased,volume was atrophied,the number of follicle were decreased,follicle volume is not consistent,the number of granular layer cell were decreased,adipose tissue hyperplasia granulose.In the group B,the number of luteal cells were decreased,volume was atrophied,the number of follicle were decreased,follicle volume is not consistent,the number of granular layer cell were decreased,interstitial hyperplasia.In the group C,large number of luteal cells,and follicles.(4)Expression of SRB1 and CYP2E1:The expression of CYP2E1 and SRB1 in A group and B group were significantly higher than group N1(P<0.05), the expression of SRB1 and CYP2E1 in group C were significantly higher than group N2(P< 0.05).There was no significant difference in expression of SRB1 and CYP2E1 between the group A and the group B (P> 0.05),but here was significant difference in the expression of SRB1 and CYP2E1 between the group A and the group C (P<0.05). Conclusion:POF group was most similar to the definition of POF disease,abnormal sapra was more easily causes POF disease than abnormal sawda.
