Effects Of Ozone Treatment On IL-1β ã€TNF-α And VEGF In Serum And Retinal Tissue Of Diabetic Rats | | Posted on:2017-02-21 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Lou | Full Text:PDF | | GTID:2284330485451312 | Subject:Ophthalmology | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the effect of ozone oninterleukin-1β ã€tumor necrosis factor-α ã€vascular endothelial growth factor of diabetic retinopathy in rats induced by STZ.Methods: 60 male rats,16 rats were selected as the controlgroup, 44 rats were injected intraperitoneally with streptozotocin(STZ,40mg/kg) after high glucose and high fat diet. 40 successful models were randomly divided into diabetic rats group and ozone therapy group,each has 20 rats.ozone therapy group treated with ozone enema 3 times a day for 8 weeks. diabetic rats groupgot no treatment.The weight and blood glucose of rats were monitored regularly.The eyeball was drawn and the blood was taken from abdominal aorta after 8 weeks.Evans blue angiography in the detection of retinal conditions. The content of nterleukin-1 β 〠tumor necrosis factor- α in retina was detected by immunohistochemistry.The content of interleukin-1β ã€tumor necrosis factor-α ã€vascular endothelial growth factor in serum and retina was detected by enzyme linked immunosorbent assay.Results:(1)The blood glucose in the group of diabetic rats and ozone therapy was higher than control group.The body weight were both lower than control group,the result was significant difference(P<0.05). there was no significant difference when compared each other(P>0.05).(2)Evans blue angiography showed no significant changes in retinal vascular morphology in control group. In diabetic rats group and ozone therapy group,fluorescence leakage, shallow and deep veins became dense, circuity.The retinal vascular in two groups were higher than control group, while retinal vascular in ozone therapy group was reduced compared to diabetic rats group,there was a statistical significance(P <0.05).(3)The expression levels of interleukin-1 β 〠tumor necrosis factor-α in RGCs of ozone therapy group were lower than diabetic rats group but higher than control group, there was a statistical significance(P <0.05).(4)The levels of three factorsin Serum and retinal tissue in diabetic rats group and ozone therapy group were higher than control group and the expression of three factors inozone therapy group was lower than diabetic rats group(P< 0.05).Conclusion:Ozone therapy can improve diabetic rats retinal vascular rate and vascular leakage, and reduce the retinal tissue and interleukin-1 β 〠tumor necrosis factor- α 〠vascular endothelial growth factor expression,suggesting that ozone therapy may play a role in delaying the development of DR in the early stage. | | Keywords/Search Tags: | O zone, inflammatory response, interleukin-1β, tumor necrosis factor-α, vascular endothelial growth factor, diabetic retinopathy | PDF Full Text Request | Related items |
