Objective To investigate the effect of autologous platelet rich plasma(PRP) in tendon bone healing after reconstruction of the anterior cruciate ligament(ACL).Methods 30 health New Zealand white rabbits, aged 3-4 months, weight in line,there were no obvious limbs athlete’s foot after physical examination,Thirty New Zealand rabbits were divided into the experimental group and the control group randomly,each group of 15 only.. In the experimental group, the platelet rich plasma was obtained by the method of two-step centrifugation, and the experimental group was implanted with PRP and fibrin gel. 30 New Zealand white rabbit will adopt flip a coin to send randomly on one side of the knee anterior cruciate ligament injury model was established,The control group only transplant into biological protein gel. At 4, 8, 12 weeks, 5 rabbits were sacrificed at 3 time points, and 3 were used for bone graft versus tension test, independent sample t test was used. Two for the pathological observation, On two groups of specimens after dyeing BUARK score, using Wilcoxon rank sum test, observe the healing of tendon bone healing after ACL reconstruction.Results After 4 weeks: the control group still visible large necrotic tissue, only a small amount of fibroblasts, no obvious collagen fiber, and fiber arrangement disorder. PRP group of necrotic tissue is relatively small, is a small amount of fibroblasts and collagen fibers begin to see the.cause, organizations still disordered arrangement. Postoperative 8 weeks: the control group is a large number of fibroblasts, tendon bone healing interface has a small amount of collagen fiber.cause, fiber arrangement still owe rules. PRP group is a large number of fibroblasts, tendon bone healing interface has a large number of collagen fibers last, most of the fiber began to appear orderly arrangement, but there are still a small amount of fiber disorderly growth. 12 weeks postoperatively, less control of fiber cells, and increased formation of collagen fibers, fibroblasts and collagen fibers in tendon bone healing at the junction of distribution. PRP of fiber cells decreased significantly, the residual only a small amount of fibroblasts, rules of visible tendon bone healing at the junction of a large number of collagen fibers. Graft versus pull experiment showed that the experimental group and control group of the femur, graft is the pull strength and time length, the experimental group and control group 4 weeks after the pull strength compared to no obvious statistical significance(P > 0.01), from 8 weeks to 12 weeks of the PRP group significantly better than the control group and the difference was statistically significant(P < 0.01).Conclusion PRP can promote the formation of collagen fibers and the mechanical strength of the tendon bone interface.
