| Purposes:To investigate the expression of pericyte in different regions of salivary adenoid cystic carcinomas, and its relationship with salivary adenoid cystic carcinoma clinical parameters.Methods:With the method of immunohistochemical staining, the pericytes of salivary adenoid cystic carcinoma specimens were stained with anti-NG-2 antibody. First of all, among all 50 cases, each specimen was divided into two subgroups, respectively was: the tumor center area and the tumor peripheral area. Firstly, at low magnification, find four regions with high density of blood vessels(each subgroup had two regions), then, turn to the high magnification, each microscope unit vision was measured with microvessel density (MVD), and microvessel pericyte density (MPD),so as to calculate the microvessel pericyte coverage index (MPI)., then averaged. According to the different clinical characteristics of patients (age, gender, tumor location, tumor size, tumor invasion and metastases, clinical stage), specimens were divided into groups, and statistical analysis were carried out among different groups for detecting microvessel density (MVD), microvessel pericyte density (MPD), microvessel pericyte coverage index (MPI).Results:(1)The expression of MVD among women (6.075±1.347) was higher than men (5.100±1.370) with statistical significance, P= 0.047. The expression of MVD in tumor peripheral area(6.280±1.738) is higher than the tumor central area(5.480± 1.418), P= 0.013. Otherwise, MVD had a positive correlation with the tumor invasion and metastasis, the expression of MVD among tumors with invasion and metastasis (6.233±1.357) was higher than those without invasion and metastasis (5.350±1.308), P= 0.027.In another different groups of patients, the expression of MVD had no obvious statistical significance.(2) The expression of MPD in tumor central area (2.460±1.249) was higher than the tumor peripheral area (1.860±0.833) with statistical significance, P= 0.006. MPD had a negative correlation with the tumor invasion and metastasis, and its expression in tumors without invasion and metastasis(2.450±0.826) was higher than those with that(1.967±0.765), P= 0.039. In another different groups of patients, the expression of MPD had no obvious statistical significance.(3) MPI varies among different tumor areas, the central area (0.456±0.225) was higher than the tumor peripheral area (0.320±0.179), P= 0.001. MPI had a negative correlation with the tumor invasion and metastasis, and its expression in tumors without invasion and metastasis(0.462±0.122) was higher than those with invasion and metastasis(0.317±0.101), P= 0.000. In another different groups of patients, MPI had no obvious statistical significance.Conclusions:The expression of pericyte is diffenent among different areas in salivary adenoid cystic carcinoma.Microvessel pericyte coverage index has a negative correlation with the tumor invasion and metastasis. Pericyte plays an important role in tumor invasion and metastasis of salivary adenoid cystic carcinomaSignifiance:This experimental study found that pericyte plays an important role in salivary adenoid cystic carcinoma, thus we can take pericyte as a new treatment target, and it provides a new insight in clinical treatment of salivary adenoid cystic carcinoma. Meanwhile, the microvessel density has a close relationship with tumorigenesis, local invasion and distant metastasis, thus, we can take the capillaries as a therapeutic target, and it can reduce capillary number, inhibit the growth of tumor, maybe it could be another kind of new treatments. |
