Objectives:1. Detection of the expression state of microRNA-203 in cervical cervical lesion tissue, discuss its role in the development of cervical lesions.2. Culture the cervical cancer cells under different concentration of folic acid interfere, detect the expression state of microRNA-203 methylation and the expression level of microRNA-203. evaluate the role of folate deficiency in the development of cervical cancer.Methods:1. Source of specimen:we randomly collected the 90 patients treated with colposcopic examinations from 2012 January to 2013 January. According to the results of cervical biopsy pathologic diagnosis under colposcopy. divided into cervical intraepithelial neoplasia in 30 cases, squamous cell carcinoma (SCC) in 30 cases as study group; the other with negative for intraepithelial lesion or malignancy as the control group of 30 cases.2. Detected the miR-203 expression level of cervical tissue by RT-PCR technique.3. Regularly cultured Caski cervical cancer cell lines, gave the different concentrations of folic acid, methylation specific PCR (MSP) method to detect microRNA-203 methylation status, RT-PCR method to detect the expression level of microRNA-203 in cervical cancer cells.Results:1. IN normal cervical tissue、CIN, cervical cancer tissue, relative expression of microRNA-203 were 1.00±0.10,0.13±0.03,0.27±0.05. Each group of microRNA-203 expression quantity was using single factor analysis of variance, microRNA-203 expression difference was statistically significant (F= 6.414. P= 6.414). Each group of mieroRNA-203 expression of group comparison was using the LSD-t test. microRNA-203 between normal group and CIN. the expression difference was statistically significant (P= 0.001). and the cervical cancer group and normal group CIN and cervical cancer group expression of microRNA-203 had no statistically significant difference (P= 0.151. P=0.060).2. In 1.0 u g/ml. 10.0u g/ml,100.0 u g/ml.250 u g/ml.750 u g/ml.1000.0 u g/ml folic acid of different concentration levels of microRNA-203 CpG island methylation rate were 60.00%,53.33%,40.00%.33.33%.13.33%.10.00%. with a higher level of folic acid, the methylation rate was on the decline (X2=27.473, P< 0.001). the expression of miR-203 levels was on the rise (F=427.416, P<0.001). Analysized the correlation of CpG island methylation rate of microRNA-203 and the expression level of microRNA-203, a negative correlation (rs= 0.886. P= 0.019).Conclusions:1. The cervical lesion tissues compared to normal cervical tissue, the expression levels of miR-203 was downregulated. the downregulated of miR-203 expression plays an important role on cervical cancer happening.2. Lack of folic acid may result to high methylation of miR-203 and miR-203 lower expression,which are important events during the cervical lesions occurred. Folic acid supplements may prevent the occurrence of cervical cancer and precancerous lesions in the further development of cervical cancer.
