Effect And Mechanism Research On Three-Dimensional Multicellular Spheroids Models Of Colon And Lung Cancer Of Anthricin Combined With Fluorouracil

Objective:To investigate the effects of Anthricin combined with fluorouracil and the mechanism onSW480,SW620 and A549 three-dimensional multicellular spheroids models,providing the basic research data for further elucidate the anti-tumor mechanism of Anthriscus sylvestris.Methods:Establishing the SW480,SW620 and A549 three dimensional multi-cellular spheroids model,testing Anthricin combined with fluorouracil in the inhibition effects on these three kind of three-dimensional multicellular spheroids models by CCK8 method;Testing Anthricin combined with fluorouracil in the influnences of the cell cycle and apoptosis on these three kind of three-dimensional multicellular spheroids modles by the flow cytometry;Detecting the influences of Anthricin combined with fluorouracilinexpressiones of FAK and COX2 protein in these three kind ofthree-dimensional multicellular spheroidsmodles.Results: Anthricin could obviously inhibit SW480,SW620 and A549 three-dimensional multicellular spheroids models and it showed a certain dose-dependent,the ICsowere 26.82μg/ml,34.21μg/ml,47.92μg/ml; Anthricin combined with fluorouracil could significantly decrease the activity of these three kind ofthree-dimensional multicellular spheroids modles,and the activity would decrease with the concentration’s increase of Anthricin;5-FU could make these three kind of cells’cycle be arrested in S phase and Anthricin could make their cell cycle be arrested in S phase and G2/M phase, Anthricin combined with fluorouracil could make these three cells’ cycle be arrested in S phase and G2/M phase and ultimately induce cell apoptosis,and they could obviously inhibit the expression of FAK and COX2 in these three kind of cells.Conclusion:Anthricin can significantly reduce the viability of SW480, SW620 and A549 two-dimensional multicellular and three-dimensional multicellular spheroids,and the effects show a certain dose-dependent; Anthricin combined with fluorouracilcan significantly reduce the viability of these three kind of multicellular spheroids, and the activity would decrease with the concentration’s increase of Anthricin.Bkocking cell cycle,Inducing cell apoptosis and regulating the signaling pathway maybe one of the inhibition echanisms of Anthricin on these three kind of cells.
