Objective:The study is aimed to establish a model of rabbit bone of knee arthritic,which is similar to that of human knee osteoarthritis,and based on the success of this model, observe the effect of modified Youguiyin on cartilage injury of rabbit knee osteoarthritis and the expression of apoptotic genes of Fas, Fas L and P53.In order to discuss the effect of modified youguiyin in in treating knee osteoarthritis.Method: Experimental animals and groups:Choose 48 Japanese big ear rabbits and randomly divided them into 4 groups,12 rats in each group:The normal group, model group, celecoxib group and modified Youguiyin group.Modeling method:The model group, celecoxib group and Youguiyin group with medical polymer materials fixed rabbit left knee in extension for 4 weeks,to establish an experimental model of osteoarthritis of the knee in rabbits.Beginning to take medicine after the first day we determined the modeling success.The rabbits in each group were fed according to the feed rate of 250g/1 rabbit / day,in order to ensure the celecoxib group and modified Youguiyin group rabbits daily dosage.Administration intervention for 6 weeks,respectively,in which the normal group and model group were not given drug, feed the standard feed;Celecoxib group were fed with celecoxib feed(75mg/ / day by celecoxib in standard feed);Modified Youguiyin group to feed the right to drink from the right to drink feed(according to 120g/ only / day mix and add and subtract right to drink fine powder in the standard feed).After six weeks of continuous administration,the rabbits were sacrificed by ear vein air embolism method,get the experimental rabbit left knee joint cartilage surface(in order to reduce the difference of specimens,all samples are taken from experimental rabbit knee femoral condyle articular cartilage).The gross observation, light microscopy, electron microscopy and apoptosis related genes FAS, Fas L, P53 expression were detected in all the groups.Result:After drug intervention, both in the general observation, the results of light microscopy and electron microscopy, or c DNA microarray test results show subtraction Youguiyin group and celecoxib group cartilage samples were the same as the model group were significantly different but at the time of addition and subtraction Youguiyin group and celecoxib group morphological comparison, both showed no significant differences in gene expression profiling microarray results suggest that the addition and subtraction of traditional Chinese medicine and western medicine Youguiyin celecoxib having a difference in the inhibition of apoptosis cartilage path.Conclusion:(1) Compared with the normal group and the normal group, the Mankin ’s score of the normal group was compared with the model group, which showed that the model of the rabbit knee osteoarthritis model was successful;(2) Gross observation,were observed by light microscope and electron microscope were used to observe the combined method can the apoptosis of cartilage specimens in each group qualitative determination by scoring results were compared to the degree of injury was performed a quantitative analysis, to compensate for the lack of various method of observation of specimens of single observation;(3) The plus and minus right return drinking group and model group were compared, both specimens of general effect score comparison or light microscopy results Mankin’s score comparison, or is the specimen under the electron microscope observation results showed modified Yougui drink on rabbit knee osteoarthritis cartilage degeneration has a good therapeutic effect.(4) Contrast modified Yougui Yin group and celecoxib group general effect score, results Mankin’s score and electron microscope observation of observed by light microscopy, did not find significant differences, that addition and subtraction of Chinese medicine Youguiyin can reach and celecoxib the same therapeutic effect in the treatment of osteoarthritis of knee joint;(5) This study showed FAS, Fas L and p53 apoptosis factor with osteoarthritis of the knee lesions are closely related to the extent and in the set of data was positively related to the factor related apoptosis expression or with OA pathogenesis, suggesting that through the application of drugs inhibit the expression of factor related apoptosis or of OA an important role in preventing and treating;(6)Western medicine of celecoxib and addition and subtraction of Youguiyin showed inhibitory effect on chondrocyte apoptosis related factor, suggesting that the two drugs in OA treatment process plays an important role, on this basis to carry out further clinical and basic research, to study the control mechanism.(7) In this group of data, addition and subtraction of Chinese medicine right return Decoction and Western medicine celecoxib has difference in inhibiting the apoptosis of cartilage cells path, or to make in-depth study to further reveal the addition and subtraction of Chinese medicine of Youguiyin on osteoarthritis intervention mechanism.
