| Objective:To explore efficacy of TaoHeChengQiTang on CRF (Chronic Renal Failure) by study its effect on chronic renal failure rats renal function, renal pathology and TGF-B/Smads signaling pathway.Methods: â‘ Grouping and molding:Randomly divide (allocated) 60 clean female SD 7-years-old rats into 5 groups:blank group, sham-operated group, model group, control group, treatment group; 5/6 Kidney resection to rats in model groupã€control group and treatment group; Peeling kidney capsule to rats insham-operated group; no molding to blank group. â‘¡ Therapy and dose:Medical preparation was treated 28 days after molding:blank group, model group, sham-operated group was fed with distilled water; control group was fed with Uremic Clearance; treatment group was fed with TaoHeChengQiTang, treatment lasts 8 weeks.â‘¢Test index after treatment, rats’blood was collected for examination of blood routine, kidney function and serum; ruine samples over 24 hours was collected for examination of 24-hour urinary protein, β2-MG, NAG and RBP of urine. Immunohistochemical examination was adopted for the expression of renal tissue’s TGF-β1ã€Smad2ã€Smad3ã€Smad4ã€Smad7 protein, and residual renal tissue was observed under light microscope to sum up pathological morphology changes.Results: â‘ RBC, HB in model group were significantly lower than those in blank group, Scr, BUN, (32-MG of serum,24-hour urinary protein, β2-MG ofã€NAG and RBPof urine were higher (P<0.05), TGF-β1ã€Smad2ã€Smad3 in renal tissue were higher.. and Smad4 and Smad7 lower (P<0.05).â‘¡ RBCã€HB in treatment group and control group were significantly higher than those in model group. Scr. BUN. β2-MG of serum,24 hour urinary protein,β2-MGã€NAG and RBP of urine were lower (P<0.05), TGF-β1ã€Smad2〠Smad3 in renal tissue were lower, and Smad4 and Smad7 higher (P<0.05). â‘¢In treatment group and control group Scr, BUN, β2-MG of serum.24-hour urinary protein,. β2-MGã€NAG and RBP of urine were lower (P<0.05), TGF-β1ã€Smad2ã€Smad3 in renal tissue were lower. and Smad4 and Smad7 higher (P<0.05). â‘£Renal tissue pathological observation:in normal group, there was no pathological changes in glomerulus and kidney tubules; in sham-operated group, inflammatory cell was infiltrated; in model group, renal corpuscle was decreased, disorganization and congestion, hyperplasia of stroma, cyst cavity expansion, and infiltration and hyalinization of inflammatory cell was observed in glomerulus. tubular ectasia was observed and tubular epithelia was swelled; in control group, the moderate congestion and expansion of cyst cavity was observed in glomerulus, tubular ectasia was observed, the convoluted tubule epithelia was swelling and small amount of inflammatory cells was infiltrated in stroma; in treatment group, glomerulus was slightly congested, cyst cavity was not expanded, some of the inflammatory cells was infiltrated in stroma, and tubular was as normal.Conclusions: â‘ CRF rat’s renal function can be significantly improved by TaoHeChengQi Tang, and Scrã€BUNã€Î²2-MG of serum can also be decreased. â‘¡TaoHeChengQiTang is good to protect tubular and decrease 24-hour urinary protein,β2-MGã€NAG and RBPof urine. â‘¢Chronic renal failure can be delayed by TaoHeChengQiTang, it would probably achieve kidney interstitial fibrosis by producing influence onTGF-β/Smads signaling pathway to inhibite TGF-β1ã€Smad2ã€Smad3 and promote Smad4ã€Smad7. |
