| Various kinds of small molecule unsaturated fatty acid have been demonstrated to benefit human health. 2-dodecenoic acid(BDSF, a diffusible signal factor from Burkholderia cenocepacia), which is capable of inhibiting the dimorphic switching and block the formation of biofilm of Candida albicans in previous study, has attracted great interest. In order to further explore the clinic potential of BDSF, we investigated the effect of BDSF in wound healing as follows.1. The effect of BDSF on the skin wound healing in the back of SD rats was first studied. ELISA(enzyme-linked immune sorbent assay) were applied to estimate inflammatory cell factors, which indicated the healing status of the skin wound. Compared with murine model treated with PBS, interleukin 1 and interleukin 6 were repressed by BDSF, suggesting 300 μM of BDSF accelerated the wound healing significantly. Moreover, ELISA, HE(hematoxylin and eosin) staining biopsy, and immunohistochemical stain showed that BDSF increased the vascular endothelial growth factor, and promoted the thickening of the epidermis layer, inward extending, and closely packing of internal organization. In one word, BDSF could expedite the wound healing through facilitating the formation of blood vessels and epithelization.2. The repair ability of BDSF on HEK-a cell damaged by Candida albicans was researched. 44 cytokines were measured by cytokines chip. Monocyte chemotactic factor 1 and 2(MCP-1 and MCP-2), restructuring chemokine(I-309), granulocyte colony stimulating factor(GCSF), growth related factor(GRO), and mast cell growth factor(SCF) were decreased by BDSF obviously, indicating that BDSF could promote the repair of the cell damage caused by Candida albicans.3. The tumor suppressor ability of BDSF was estimated in A549 cells and murine lung cancer model. Flow cytometry(FCM) and PCNA kit were used to state the effect of BDSF on the apoptosis and proliferation of A549 cells, respectively. BDSF was given to nude mice carrying lung cancer by gavage, and several kinds of serous cytokines of were measured. Both experiments showed that BDSF did not affect the growth of cancer cells. |
