ObjectiveDendrobium officinale Kimura et Migo is a kind of precious tradition Chinese medicine, it can strengthen stomach, promote fluid production, nourish Yin and clear heat. Dendrobium used for medicine can be traced back to the Han dynasty and dendrobium officinale is one of the highest value traditional Chinese medicine of it. As people’s living standards improve, more and more people began to pay attention to health, Dendrobium officinale as "the first nine immortality" getting into the view of ordinary people. Mostly, dendrobium officinale used for medicine was decoction and also used to make soup Edible. There were many chemical composition inside the dendrobium officinal and related scholars had done a lot of research. However, the water-soluble small molecule compounds were rarely reported. We had done the research about the difference between the stems and the leaves of dendrobium officinale about the compounds. The research showed that the species of the compounds in the stems was same as the leaves of demdrobium officinale. Therefore, The article was based on the research we had done before and started from the decoction of dendrobium offieinale leaves, Isolated and purified the soluble monomer compounds from them. The activity of water-soluble monomer compounds on spleen lymphocyte proliferation in mice was screened. We hope to provide the basis for the study of chemical substances of dendrobium officinale; providing a scientific basis for the use of dendrobium officinale leaves.Method:1. Dendrobium officinale was extracted with water. Water-soluble components was seprated by using AB-8, Sephadex LH-20, ODS and preparative HPLC. The water-soluble components’structure were identificated by using modern spectroscopy like HPLC, ESI-MS,1H-NMR,13C-NMR.2. MTT method was used to screen the activity on spleen lymphocyte proliferation in mice.Results:1. Structures of the isolated six water-soluble compounds was identified, namely:rutin (1), isovitexin-2"-O-β-D-glucopyranoside (2), Isovitexin (3), Vitexin-4"-O-glucoside (4), apigenin-6-C- β -L-arabinopyranosyl-8-C- β-D-glucopyranoside (5), a new compound was also separated and named as bioside temporarily.2. Screening result of the activity on spleen lymphocyte cell in mice showed compound 2,5,6has certain proliferation effect on the spleen lymphocyte cell in mice.Conclusion:The water-soluble of Dendrobium officinale leaves has many flavonoid glycoside compounds. Compound 2,5,6 isolated from dendrobium officinale leaves has certain proliferation effect on the spleen lymphocyte cell in mice. But the proliferation effect is not obvious.
