| Objective:This study was designed to investigate pipeline on human gastric cancer cell line BGC-823 inhibition and apoptosis induction mechanisms;Research methods:①by MTT assay to detect inhibition of proliferation of piperine acting on cultured human gastric cancer cell line BGC-823 at different concentrations with its time correlation;② Through the scratch test method,detect the influence of piperine on cultured human gastric cancer cell line BGC-823 migration ability and its time correlation;③ Through Transwell assay to detect the people piperine on cultured gastric cancer cell line BGC-823 invasive ability and its time correlation;④ Flow cytometry was used to detect apoptosis of different concentrations of piperine on human gastric cancer cell line BGC-823 and its concentration correlation;⑤ By assay of rhodamine to detect the influence of piperine acting on human gastric cancer cell line BGC-823 potential s of and its concentration correlation;⑥ By assay of RT-PCR to detect related gene’s expression changes of piperine acting on human gastric cancer cell line BGC-823 in mitochondrial apoptosis pathway and its concentrations correlation;⑦ Trough western in mitochondrial apoptotic pathway related protein expression interventions of piperine acting on human gastric cancer cell line BGC-823and its concentrations correlation.Research result:①ifferent concentrations of piperine acting on human gastric cancer cell line BGC-823 cells for 24h or 48h, the proliferation ability of cells was significantly reduced,with concentration-dependent and time-dependent;② Piperine acting on human gastric cancer cell line BGC-823 cells for 24h or 48h, human gastric cancer cell line BGC-823 cell migration was significantly decreased,with concentration-dependent manner;③ Piperine acting on human gastric cancer cell line BGC-823 cells for 24h or 48h, through transwell chambers cell number was significantly reduced compared with the control group,and its invasion inhibition rate showed a concentration-dependent manner;④ Piperine acting on human gastric cancer cell line BGC-823 cells for 24h, obvious early apoptotic cells appeard in a concentration-dependent manner;⑤ Piperine acting on human gastric cancer cell line BGC-823 cells for 24h, mitochondrial potential decreased in a concentration-dependent manner;⑥ Piperine acting on human gastric cancer cell line BGC-823 cells for 24h, mitochondrial pathway of apoptosis-related genes Bax, Bak with piperine concentration, the higher the expression level; Bcl-2, Survivin with piperine concentration, the lower the expression level;⑦ Piperine acting on human gastric cancer cell line BGC-823 cells for 24h, mitochondrial apoptosis pathway proteins Bax, cleaved-caspase 9 with piperine concentration increased, the higher the expression leveled; Bcl-2, and with piperine concentration increased, the lower the expression leveled.Research conclusion:Piperine could inhibit the proliferation of BGC-823 human gastric cancer cell lines, migration and invasion; piperine can induce human gastric cancer cell line BGC-823 cell apoptosis; piperine BGC-823 induced apoptosis of gastric cancer cell lines possibly through mitochondrial signaling pathway implemented. |
